Transcriptomic and Ectoine Analysis of Halotolerant Nocardiopsis gilva YIM 90087T Under Salt Stress

Abstract: The genus Nocardiopsis is an unique actinobacterial group that widely distributed in hypersaline environments. In this study, we investigated the growth conditions, transcriptome analysis, production and accumulation of ectoine by Nocardiopsis gilva YIM 90087T under salt stress. The colony color of N. gilva YIM 90087T changed from yellow to white under salt stress conditions. Accumulation of ectoine and hydroxyectoine in cells was an efficient way to regulate osmotic pressure. The ectoine synthesis was studied… Show more

“…The genetic organization of the ectABC operon from the strain is in canonical arrangement (Schwibbert et al 2011;Widderich et al 2014Widderich et al , 2016) as all genes were located in the same cluster. The presence and the arrangement of these fourth genes in an operon is also found in some other halophilic bacteria such as the industrial strain H. elongata, C. salexigens, and Streptomyces coelicolor (Han et al 2018). However, a different genetic organization of the ectABCD genes in the genome sequence of P. halophilum was observed.…”

“…Ectoine is a widely distributed solute in different halophilic and halotolerant microorganisms, produced in response to osmotic stress and functions as a potent protector of osmostress [ 66 , 67 ]. Ectoine improve protein folding and protects biomolecules such as enzymes, nucleic acids, and even whole cells against various stress conditions [ 68 ]. These multifunctional effects have fostered the development of a wide range of skincare and dermatological cosmetic preparations as moisturizers in cosmetics for the care of aged, dry or irritated skin [ 69 , 70 ].…”

“…Together, these results imply that NaCl stress is a key regulator for synthesis and accumulation of ectoine and hydroxyectoine. The ectABCD genes, related to ectoines synthesis in many halophilic strains, as H. elongata or Nicardiopsis gilva, present different degrees of transcriptional regulation in the range from 5 to 15% NaCl concentration, particularly at 15% (He et al 2015;Han et al 2018).…”
