Transcriptomic analysis of chicken Myozenin 3 regulation reveals its potential role in cell proliferation

Abstract: Embryonic muscle development and fibre type differentiation has always been a topic of great importance due to its impact on both human health and farm animal financial values. Myozenin3 (Myoz3) is an important candidate gene that may regulate these processes. In the current study, we knocked down and overexpressed Myoz3 in chicken embryonic fibroblasts (CEFs) and chicken myoblasts, then utilized RNA-seq technology to screen genes, pathways and biological processes associated with Myoz3. Multiple differentiall… Show more

“…DES). The increased abundance in MYOZ3 in strain 5 WB compared to strains 2 and 3 WB indicates that strain 5 WB may have a greater muscle growth rate since MYOZ3 plays a role in cell proliferation ( Ye et al., 2017 ), which is consistent with a higher breast weight and breast yield in strain 5. Both MYOZ3 and LDB3 contribute to Z-disc formation.…”

“…Both MYOZ3 and LDB3 contribute to Z-disc formation. The lower abundance of any of these 2 proteins in WB of strains 2 and 3 compared to strain 5 may lead to aberrant Z-disc signal transduction and muscle development ( Ye et al., 2017 ). Another Z-disc protein, ACTN2 plays a role in thin filament organization and the interaction between muscle membrane and the sarcomeric cytoskeleton ( Mills et al., 2001 ).…”

“…Further, calsarcin-3 which binds to calcineurin and inhibits the NFAT activation is implicated in negative regulation of NFAM1 expression. 34 Reduced RANK expression in CD11b deficient mice suppresses NFATc1, which induces the BCL6 (transcriptional repressor B-cell lymphoma 6) to NFATc1 gene promoter and downstream effectors beta3 integrin and cathepsin K. 35,36 We identified that inhibition of NFAM1 suppresses NFATc1 but not NFATc2 transcription factor expression during OCL differentiation. Since the antibodies detected multiple products of NFAT and OCL being a multinucleated cell, the effect of NFAM1-KD could influence the levels of NFATc1 nuclear localization.…”

“…MYOZ3 belongs to the myozenin family, of which three other members were previously proposed as tenderness biomarkers [ 13 ]. MYOZ3 acts as an intracellular binding protein to link with Z-disc proteins such as alpha-actinin and gamma-filamin and transmit calcineurin signalling to the sarcomere [ 27 ]. Due to the capacity to bind multiple proteins, the relationship between MYOZ3 and meat quality, specifically tenderness, could be through regulating the Z-disc structure and signal transduction, influencing muscle fibre differentiation [ 28 ].…”
