ABSTRACTDormancy is one of the most important adaptive mechanisms developed by perennial plants, in order to survive the low temperatures of autumn and winter in temperate climates. The study of the genes regulated during dormancy release is crucial to understand the process, with the final objective of the development of new varieties with a better adaptation to certain environments; and this is particularly important considering the increasing economical weight of fruit crops in low and medium chilling regions as the Mediterranean area. We focused on the molecular and physiological mechanisms underlying the maintenance and release of seasonal dormancy in peach. In order to achieve this we first used suppression subtractive hybridization (SSH) to identify genes expressed in dormant and dormancy-released buds in two cultivars with different chilling requirements, 'Zincal-5' and 'Springlady', and subsequently validated their differential expression utilizing a peach cDNA microarray platform containing transcripts enriched in flower buds. Additionally, we carried out a genome-wide search of peach genes related to dormancy release by hybridizing the previous cDNA microarray with mRNA samples from 10 cultivars showing different dormancy behaviour, followed by an expression correlation analysis.Among the most relevant genes identified in these two first works, we found the DORMANCY ASSOCIATED MADS-box genes DAM4, DAM5 and DAM6, described independently by other groups working in peach and other species. The central role of DAM genes in dormancy regulation has also been confirmed by additional functional approaches as the analysis of the non-dormant evg mutant, QTL analysis, and transgenic approaches.In our second work we focused on the molecular mechanisms of DAM6 down-regulation concomitant with dormancy release in flower buds. A ChIP analysis of DAM6 promoter and structural gene revealed chromatin modification events similar to those observed in vernalization of Arabidopsis and cereals. We showed that DAM6 is transcriptionally active in dormant buds collected in October, when a short chromatin region around its ATG was trimethylated in histone H3 at K4 (H3K4) and acetylated at the N-terminal tail of H3. Concomitantly with DAM6 repression, H3K4 became demethylated and H3 deacetylated. Later H3K27 was found trimethylated along a genomic region larger than 4kb, including promoter, coding sequence and intron. Effects of Fluridone Treatment on Seed Germination and Dormancy-associated Gene Expression in an Ornamental Peach (<i>Prunus persica</i> (L.) Batsch)

Fluridone, an inhibitor of carotenoid biosynthesis, is known to prevent abscisic acid (ABA) biosynthesis and to affect the germination and dormancy of seeds in several plant species. This study investigated the effects of fluridone on seed germination of the ornamental peach 'Yaguchi', and on the transcript levels of genes related to seed dormancy in this plant. Seeds were rinsed for 2 days under running tap water (RS), then soaked for 1 day in 100 μM fluridone (F), and kept at 5°C for 2 weeks (2W). The germination rate significantly increased from 0% to 9.5% in the RS+F treatment and to 71.4% in the RS+F+2W treatment, while no germination occurred in the RS and RS+2W treatments. Seedlings in RS+F and RS+F+2W treatments formed dwarf shoots, i.e. about 10 cm, and morphological lesions on the leaves. The ABA content in embryonic axes decreased after RS and increased with RS+2W. It was decreased by the RS+F+2W treatment. RS+F+2W downregulated ABA-hy3, which encodes enzymes with key roles in ABA catabolism, while its effects on 9-cisepoxycarotenoid dioxygenase (NCED1) in relation to ABA synthesis fluctuated. In contrast, Empfindlicherim Dunkelroten Licht 1 (EID1) was upregulated after fluridone treatment, suggesting that fluridone may activate positive ABA signaling pathways. Expression of GA2-oxidase8 (GA2-ox8) was not affected by fluridone. D-34) were downregulated in the RS+F+2W treatment, suggesting that the expression of these genes are controlled by low temperature and the ABA inhibitor, and are involved in seed dormancy. These results suggest that ABA inhibitor treatments can be an alternative method to promote germination by controlling ABA content and its metabolism, and consequently changing expression of certain ABA-and dormancy-related genes including ABA-hy3, EID1, LeMADS, and LEA D-34, even under insufficient chilling conditions. Plant development is orchestrated and yet flexible. This enables the formation of the correct organs within the context of previous and current environmental cues. The use of previous environmental experiences, such as the conditions when the seed formed or whether winter has passed, is critical to maximise reproductive success. Temperature is a crucial signal in achieving this, it provides current seasonal information and is an indicator of forthcoming environmental conditions, including drought or freezing. Therefore, many temperature responses facilitate environmental acclimation as well as trigger key developmental changes, such as germination, growth, and flowering. Research has identified that temperature signals are interpreted in the context of light signalling, circadian and light intensity, and developmental stage. Furthermore, a number of temperature cues are stress signals, associated with very high or low temperatures. These signals do trigger developmental responses but not as part of standard developmental growth. This article focuses on the role of ambient temperature on each of the major stages of plant development.
