Transcriptome Analyses of Two Citrus Cultivars (Shiranuhi and Huangguogan) in Seedling Etiolation

Abstract: Citrus species are among the most important fruit crops. However, gene regulation and signaling pathways related to etiolation in this crop remain unknown. Using Illumina sequencing technology, modification of global gene expression in two hybrid citrus cultivars—Huangguogan and Shiranuhi, respectively—were investigated. More than 834.16 million clean reads and 125.12 Gb of RNA-seq data were obtained, more than 91.37% reads had a quality score of Q30. 124,952 unigenes were finally generated with a mean length … Show more

“…The germination of seeds from Citrus genotypes was reported to be generally high when seeds are sown immediately after fruit harvest [7][8][9][10][11], although it may differ among species and genotypes [10]. This fact is hypothesized to be due to excessive desiccation, which might cause membrane disruption [7].…”

“…Moreover, their low and slow germination may represent a main limitation for sprouting purposes, especially for homemade production, where the consumer requires a feasible, short, and cheap way to obtain sprouts. However, the germination of Citrus species is reported to be high when seeds are sown immediately after separation from the fruit pulp [7][8][9][10][11]. Finally, the germination performance and phytochemical content of Citrus sprouts is expected to depend on the genotype.…”

“…We recently analyzed the transcriptome of etiolated citrus seedlings, there were variegated and green seedlings in both Shiranuhi and Huangguogan under the same conditions of seedling germination 23 . Although variegated plants exist widely in nature, the mechanism of citrus leaf variegation and the mechanisms regulating the expression patterns of well-characterized genes involved in photosynthesis and chlorophyll biosynthesis are still unknown.…”

“…Huangguogan (Citrus reticulata × Citrus sinensis) is hybrid citrus cultivars, which have been identified as a new cultivated variety in China. In recent years, the plant area of Huangguogan has been expanded rapidly in the southwest of China [2]. Huangguogan was a hybrid variety of Rutaceae Citrus family.…”
