Transcriptional repression of IKKβ by p53 in arsenite-induced GADD45α accumulation and apoptosis

Abstract: Our previous studies revealed that GADD45α is a liable protein, which undergoes MDM2-dependent constitutive ubiquitination and degradation in resting HepG2 hepatoma cells. Arsenite exposure induces ribosomal stress responses mediated by the ribosomal protein S7, which can block MDM2 activity and result in GADD45α accumulation and cell apoptosis. In the present study, we found that one of the catalytic subunits of IκB kinase (IKK), IKKβ, exerted a novel IKKα- and NF-κB-independent function in stabilizing MDM2 a… Show more

“…In addition, the stability of crucial factors in regulatory pathways may affect signal transduction. IKBKB could promote the stability of MDM2 ( 42 ), which can interact with MAPSK3 to promote STAT3 degradation ( 43 ). In conclusion, the proteins we screened have critical functions in cellular biological processes and lipid metabolism and may modulate the adipocytokine signaling pathway in a synergistic mode.…”

“…Ubiquitination, an important event for protein relocation and degradation, is emerging as a new mechanism for immune regulation [ 28 ]. In the present study, in despite the fact that the loading of endotoxin-containing pathogen facilitate the formation of Akt+ IKKα/β+ Rab5+ signalosome, the exact mechanisms that the endosomal relocation of Akt and IKK promoting endotoxin-containing pathogen-derived cross-presentation are still to be clarified.…”

“…The proteins encoding by PER2, GADD45A, and TPD52 genes are also involved in the cancer cell proliferation and tumor growth and are dysregulated in cancer cells (Wang et al 2016;Xiang et al 2018;Xiong et al 2018;Zhang et al 2018;Hu et al 2019;Rahman et al 2019). At the same time, the expression of these genes was resistant to the silencing of adipokine NAMPT in the glioma cells.…”
