Transcriptional repression by AML1 and LEF-1 is mediated by the TLE/Groucho corepressors

Abstract: The mammalian AML͞CBF␣ runt domain (RD) transcription factors regulate hematopoiesis and osteoblast differentiation. Like their Drosophila counterparts, most mammalian RD proteins terminate in a common pentapeptide, VWRPY, which serves to recruit the corepressor Groucho (Gro). Using a yeast two-hybrid assay, in vitro association and pull-down experiments, we demonstrate that Gro and its mammalian homolog TLE1 specifically interact with AML1 and AML2. In addition to the VWRPY motif, other C-terminal sequences a… Show more

“…Furthermore, we have preliminary data to support the idea that Grg5 is a coactivator of the transcription factor Cbfa1/Runx2. Grg5 is also reported to interact with Aml1/Cbfa2/Runx1 and modulate its transcriptional activity in vitro (Imai et al, 1998;Levanon et al, 1998;Lutterbach et al, 2000;McLarren et al, 2000). We suggest that Grg5 regulates Cbfa1/Runx2 in the growth plate and that this regulation in turn controls Ihh signaling.…”

“…Cofactors that modulate Tcf activity may also influence the effects of butyrate in HCT-116 cells. Overexpression of TLE1, a corepressor of Tcf in mammalian cells, 49 abrogated the upregulation of expression from the matrilysin promoter (p Ͻ 0.01) in butyratetreated HCT-116 cells (data not shown). However, the role of TLE in modulating Tcf4-dependent Wnt activity is controversial.…”

“…have been identified. 21,[56][57][58] Also, a synergy between b-catenin-TCF/LEF complexes and other transcription factors (including AP-1 or PEA-3) have been described in the promoter regulation of several b-catenin/TCF targets genes including MMP-7 or the g2 chain of laminin-5. 35,59 The human MCP-1 promoter has accordingly been shown to be activated by several transcription factors including NFjB, Sp1 or AP-1.…”
