Transcriptional regulation of the Saccharomyces cerevisiae amino acid permease gene BAP2

Abstract: Uptake of branched-chain amino acids by Saccharomyces cerevisiae from media containing a preferred nitrogen source is mediated by the permeases encoded by BAP2, BAP3, and VAP1/TAT1. The transcriptional activity of the BAP2 promoter is affected by a number of genes, including SSY1, which encodes an amino acid permease homologue that is necessary for transcription of BAP2. Other genes that control BAP2 encode known (Leu3p, Tup1p) and putative (Stp1p, Stp2p) transcription factors. We present evidence that the zin… Show more

“…LEU3 encodes a zinc-finger transcription factor that regulates genes involved in the biosynthesis and uptake of branched-chain amino acids (Friden and Schimmel 1988;Nielsen et al 2001). Strain BY4742 is a leucine auxotroph, and its growth depends on external leucine.…”

“…Not surprisingly, several general factors that affect the expression of SPS-sensormodulated genes have been described. These include Abf1p, Leu3p, Tup1p, Ssn6p, Uga35p, ubiquitin, and the SCF-Grr1p E3 ubiquitin ligase complex (Iraqui et al 1999;de Boer et al 2000;Bernard and André 2001b;Forsberg et al 2001;Nielsen et al 2001). The hierarchy in which these factors operate to control gene expression remains to be elucidated.…”

“…In detailed studies focused on the promoters of BAP2 and BAP3, an aminoacid-dependent upstream activating sequence (UAS aa ) was identified (de Boer et al 1998). Both Stp1p and Stp2p are able to bind DNA fragments containing the UAS aa (de Boer et al 2000;Nielsen et al 2001). Similar UAS aa motifs are present in the promoters of several other SPSsensor target genes.…”

“…This liberates the DNA binding and transactivation domains from an ϳ10-kDa N-terminal regulatory fragment. The shorter forms of Stp1 and Stp2 accumulate in the nucleus and bind to specific upstream activating sequences (UAS aa ) within SPS sensor-regulated promoters (24,25).…”
