Transcriptional regulation of parathyroid hormone-related protein promoter P2 by NF-κB in adult T-cell leukemia/lymphoma

Abstract: Parathyroid hormone-related protein (PTHrP) plays a primary role in the development of humoral hypercalcemia of malignancy (HHM) that occurs in the majority of patients with adult T-cell leukemia/lymphoma (ATLL) due to human T-cell lymphotropic virus type-1 (HTLV-1) infection. “…In adult T cell leukemia/lymphoma (ATLL) it was shown that the PTHrP promoter 2, which is upregulated in ATLL as well, contains a binding site for nuclear factor κB (NFκB), to which the subunits p50 and c-Rel bind in vivo. 35 Transient co-transfection with NFκB expression plasmids and PTHrP P2 luciferase reporterplasmid, showed that different combinations of the subunits cooperatively upregulated the PTHrP P2 promoter, while inhibition of NFκB activity reduced PTHrP P2 promoter-initiated transcription indicating a crucial role for NFκB in the regulation of the P2 in ATLL. It remains to be demonstrated if a similar mechanism can also account for the observed activation of P2 correlating with the invasiveness of our breast cancer cell line model.…”
