Transcriptional regulation of myotube fate specification and intrafusal muscle fiber morphogenesis

Abstract: Vertebrate muscle spindle stretch receptors are important for limb position sensation (proprioception) and stretch reflexes. The structurally complex stretch receptor arises from a single myotube, which is transformed into multiple intrafusal muscle fibers by sensory axon–dependent signal transduction that alters gene expression in the contacted myotubes. The sensory-derived signal transduction pathways that specify the fate of myotubes are very poorly understood. The zinc finger transcription factor, early gr… Show more

“…In support of this possibility, similar promoter analysis on the set of genes induced by overexpression of Egr3 in myotubes also did not yield significant enrichment of Egr-binding sites (Ref. 26 and data not shown), yet several directly regulated target genes have been identified and well-characterized from the array data set (26,27). Alternatively, this may indicate that a large number of down-regulated genes are indirectly regulated by Egr1 and/or Egr3.…”

“…The microinjections of the transgene were performed by the Northwestern Transgenic and Targeted Mutagenesis Core Facility, as described previously (52). Potential founder mice (which grew from the injected zygotes) were screened for insertion of the transgene by PCR analysis after DNA was obtained from their tail tip.…”

“…4). In addition, parvalbumin (Pv) immunohistochemistry was used as a wellestablished marker of proprioceptive sensory innervation to skeletal muscle stretch receptors (Albert et al, 2005;Tourtellotte et al, 2001), which was highly significantly reduced to 4.8% in Wnt1-cKO skeletal muscle compared with Wnt1-Ctl mice (n=2 mice and over 30 tissue sections analyzed per genotype, P<0.0001, Student's t-test) (Fig. 4).…”
