Transcriptional Profiling of the Iron Starvation Response in Bordetella pertussis Provides New Insights into Siderophore Utilization and Virulence Gene Expression

Abstract: Serological studies of patients with pertussis and the identification of antigenic Bordetella pertussis proteins support the hypothesis that B. pertussis perceives an iron starvation cue and expresses multiple iron source utilization systems in its natural human host environment. Furthermore, previous studies using a murine respiratory tract infection model showed that several of these B. pertussis iron systems are required for colonization and persistence and are differentially expressed over the course of in… Show more

“…The induction of (p)ppGpp accumulation by carbon starvation initially was shown in E. coli (15) and subsequently demonstrated in several other bacteria (16,(38)(39)(40)(41). It was reported that T3SS production in both B. pertussis and B. bronchiseptica was upregulated when bacteria were cultured in iron-depleted medium (8,42). We cultured strain PMK21 under iron-limited conditions to assess the role of the stringent response in the iron limitation response, but it did not grow in iron-depleted SSC medium (data not shown).…”

“…Although the genes are present and expressed as mRNA, it had been thought that B. pertussis did not produce or utilize the T3SS. Recently, the production of the T3SS proteins was discovered by analysis of fresh clinical isolates (7), and laboratory-adapted strains showed the production of the T3SS in cultures grown under iron-depleted conditions (8).…”

“…The switch from CDC237 to CDC300 also occurred by an inversion at the same boundaries in the fimH1 background, providing further evidence for reversibility. The biosynthesis and transport of alcaligin, an important siderophore produced by B. pertussis and B. bronchiseptica (49), were encoded nearby. Whether this inversion modulates alcaligin production or whether the observed parallelism reflects positive selection rather than neutral polymorphism has yet to be explored.…”

“…An in-frame nonpolar deletion in the dra locus in the B. pertussis Bp536 WT strain was constructed using a previously published allelic exchange method (25). A 378-bp MfeI-HindIII fragment spanning regions 5= to and including the first 25 codons of draD was amplified from Bp536 genomic DNA using the primers dra5AMfeI and dra3AHindIII (Table 2).…”
