Transcriptional Profiling of Rice Leaves Undergoing a Hypersensitive Response Like Reaction Induced by Xanthomonas oryzae pv. oryzae Cellulase

Abstract: A secreted cellulase of Xanthomonas oryzae pv. oryzae induces innate immune responses in rice including a hypersensitive response (HR) like reaction. Microarray analysis was conducted using RNA isolated 12 h following ClsA treatment of leaves. BLAST searches were performed for the 267 (152 up-and 115 downregulated) differentially expressed (≥2-fold) genes. A number of defense and stressresponse functions are upregulated while a number of functions involved in metabolism and transport are downregulated followin… Show more

“…Twenty-five to 30 leaf pieces covering the infiltrated zone from each treatment, LipA or buffer (10 mM potassium phosphate buffer, pH 6.6), were harvested 12 h after infiltration. RNA was isolated from each treatment and quality was assessed as described previously (Jha et al 2010). Eight micrograms of total RNA isolated from each treatment (LipA or buffer) was used for Affymetrix microarray hybridization.…”

“…Comparative analysis of rice innate immune responses induced by the cell wall degrading enzymes ClsA and LipA Rice gene expression changes following treatment with the cell wall degrading enzyme, ClsA, have been determined (Jha et al 2010). In order to compare the ClsA-and LipA-induced transcriptional changes in rice leaves, we have reanalyzed the ClsA-induced transcriptome data by retrieving the CEL files from the NCBI microarray repository, Gene Expression Omnibus (GEO) (http://www.ncbi.nlm.nih.gov/geo), accession number GSE8216.…”

“…The data was compared with the genes induced in response to sucrose starvation in rice suspension cells (Wang et al 2007) and cellulase treatment to rice leaves (Jha et al 2010). Microarray analysis has also been reported to identify genes induced during cell elongation (Kwon et al 2005) and cell wall regeneration using suspension cells in Arabidopsis (Irshad et al 2008).…”
