Transcriptional Induction of Endothelial Nitric Oxide Gene by Cyclosporine A

Abstract: We have previously shown that the immunosuppressant cyclosporine A (CsA) increases the activity, the protein level, and the steady-state levels of the mRNA of the endothelial nitric-oxide synthase (eNOS) gene in bovine aortic endothelial cells (BAEC). We have now investigated the mechanisms responsible for these effects. Preincubation with an inhibitor of RNA polymerase II abol- Whereas cyclosporine A (CsA) 1 still stands as a cornerstone therapy for immunosuppression, its use encompasses serious side effects … Show more

“…Our current study shows that IL-6, a major trigger of the APR, also decreases eNOS expression. Our work adds to the findings of others who have identified transcription factors regulating eNOS promoter transactivation, including Sp1, GATA, AP-1, NF-1, and Smad2 (26,27,(35)(36)(37).…”

“…Interestingly, a link between ILK and AP-1 has been recently described, 39,40 and this transcription factor seems to play a significant role in the modulation of eNOS mRNA expression by different stimuli. 37,41 In summary, HUVECs cultured on Col. I synthesize less NO, probably as a consequence of a decreased expression of eNOS mRNA, when compared with Col. IV-grown cells.…”

“…Whereas its hypertensive effects have been attributed to paracrine vasoconstrictors, CsA was also found to paradoxically augment NO production in vitro and in vivo. CsA treatment of endothelial cells in culture was shown to enhance eNOS expression 41,42 through transcriptional activation. 42 This effect of CsA on the transcriptional upregulation of eNOS was proposed to function through reactive oxygen intermediates, such as hydrogen peroxide, and reactive oxygen intermediateinduced AP-1 activity.…”
