Transcriptional analysis of amino acid, metal ion, vitamin and carbohydrate uptake in butanol-producing Clostridium beijerinckii NRRL B-598

Abstract: In-depth knowledge of cell metabolism and nutrient uptake mechanisms can lead to the development of a tool for improving acetone-butanol-ethanol (ABE) fermentation performance and help to overcome bottlenecks in the process, such as the high cost of substrates and low production rates. Over 300 genes potentially encoding transport of amino acids, metal ions, vitamins and carbohydrates were identified in the genome of the butanol-producing strain Clostridium beijerinckii NRRL B-598, based on similarity searches… Show more

“…These findings contributed to the modification of growth media to improve either the sporulation or the solvent formation. As shown in a previous transcriptomic study on C. beijerinckii (Vasylkivska et al, 2019), the expression of genes involved in amino acids, vitamins, and ions transport varied depending on the culture's growth stage. Based on this work, close to 600 genes, putatively involved in amino acid, ions and vitamin transport and metabolism, were identified.…”

“…Studies of the transcriptome of wild-type and mutant C. beijerinckii cultures during fermentation indicated changes in the expression of genes involved in ion- and amino acid transport at sporulation initiation. In C. beijerinckii NRRL B598, sporulation initiation was concomitant with an upregulation of the genes encoding a magnesium transporter, and an upregulation of genes encoding potassium, sodium, and iron transporters was detected during stationary phase (Vasylkivska et al 2019 ). In cultures of the asporogenous C. beijerinckii Δ spoIIE strain, the expression of genes encoding iron transporters were downregulated during stationary phase (Diallo et al 2020b ), indicating a potential role of iron in sporulation.…”

“…For the first two approaches, when EMS was used as a mutagenic agent, a spore suspension of C. beijerinckii NRRL B-598 was heated to 80 • C for 2 min, vortexed and transferred to Erlenmeyer flasks with TYA medium (Vasylkivska et al, 2019) containing 20 g/L of glucose (analytical reagent grade, PENTA, Chrudim, Czechia). The strain was cultivated in a Concept 400 anaerobic chamber (Ruskinn Technology) under a stable N 2 atmosphere at 37 • C (Figure 1, step 1).…”
