Transcription factor induction of human oligodendrocyte progenitor fate and differentiation

Abstract: Human oligodendrocyte progenitor cell (OPC) specification and differentiation occurs slowly and limits the potential for cell-based treatment of demyelinating disease. In this study, using FACS-based isolation and microarray analysis, we identified a set of transcription factors expressed by human primary CD140a + O4 + OPCs relative to CD133 + CD140a − neural stem/progenitor cells (NPCs). Among these, lentiviral overexpression of transcription factors ASCL1, SOX10, and NKX2.2 in NPCs was sufficient to induce S… Show more

“…Sox10 can restrict differentiation of neural precursor cells into the OL lineage, in part by regulating the expression of the Shh signaling pathway (Pozniak et al, 2010). Overexpression of Sox10 alone is sufficient to promote the commitment of neural precursor cells toward the OL lineage to form mature OLs (Wang et al, 2014). …”

“…This protocol has been further optimized by a various groups (Franco et al 2015;Neri et al 2010;. In a number of studies, ectopic overexpression of either Sox10 (C. D. Pozniak et al 2010a;Wang et al 2014), Olig2 (Copray et al 2006;) or both (Liu et al 2007) has been shown to facilitate or even induce NSC differentiation toward oligodendrocyte linage. However, this approach cannot provide immunologically matched cell types for cell therapy due to difficulty of obtaining adult NSCs.…”
