AIM: The present study investigated the role of redox balance, infl ammation, mitochondrial dysfunction, and apoptosis in Tramadol (Tra)-induced testicular toxicity. METHOD: Twenty-four male Wistar rats were randomly divided into either the control group or the groups receiving different doses of Tra (25, 50, and 75 mg/kg/day, i.p.) for 21 successive days. Testicular tissues were collected for oxidative stress, mitochondrial function, sperm assays and histopathological evaluation. Real-time polymerase chain reaction was performed to evaluate the markers of infl ammation and apoptosis. RESULTS: Tra caused a signifi cant reduction in the sperm count, motility and morphology, while it caused a marked increase in oxidative stress parameters. In addition, Tra induced testicular mitochondrial dysfunction due to the collapse of mitochondrial membrane potential and mitochondrial swelling. It also led to the signifi cant inhibition of anti-apoptotic Bcl-2 expression, besides a signifi cant increase in pro-apoptotic Bax expression. There was a signifi cant increase in the level of tumour necrosis factor-α, interlukin-1β and nuclear factor kappa B. Histopathological degenerative changes were observed in the testis after Tra exposure. CONCLUSIONS: The present results suggest that Tra exposure may lead to reproductive toxicity due to the loss of the antioxidant defence system, mitochondrial dysfunction, and activation of infl ammatory and apoptotic pathways (Tab. 4, Fig. 5, Ref. 63).

This work was designed to study the potential effect of kiwifruit, turmeric extract or their combination in alleviation of the liver toxicity induced by tramadol intake. Thirty rats were fed on a basal diet, six of them continued on the basal diet (negative control). The remaining rats were given tramadol daily and divided into four groups, one of them served as positive control, while the other three groups were treated daily with kiwifruit juice, turmeric water extract and their combination for four weeks. The results revealed that the positive control exhibited significant decreases in serum triglycerides, total cholesterol and HDL-c levels and a significant increase in serum LDL-c as compared to the negative control. In comparing with the positive control, the serum cholesterol, triglycerides, VLDL-c and HDL-c values increased while LDL-c value decreased significantly in the three treated groups which reached the normal values of the negative control. The findings revealed significant decreases in serum levels of ALT, AST, ALP, GGT and total bilirubin and significant increases in total protein and albumin of the treated groups in comparing with the positive control. An improvement of the antioxidant parameters was observed where the serum levels of tumor necrosis factor-α, acetylcholine esterase and total oxidative capacity decreased significantly while the levels of superoxide dismutase and total antioxidants capacity increased in all the treated groups in comparing with positive control group. It is recommended by using kiwifruit and turmeric extract in the diets of those who addicted to tramadol intake.

Aims: This study investigated the effect of vitamin E on serum liver enzymes and bilirubin concentrations in male Wistar rats treated with sildenafil citrate (SC) and a combination of SC and tramadol on the background that these drugs cause oxidative damage to the liver. Methods: Thirty male Wistar rats (180-200 g) were randomly divided into six groups (n=5) thus: control (0.2 ml vehicle – olive oil), SC-treated group (Sil) (10 mg/kg of SC), SC+tramadol-treated group (Sil+Tra) (10 mg/kg of SC and 20 mg/kg of tramadol), vitamin E-treated group (Vit E) (100mg/kg of vitamin E), SC+vitamin E-treated group (Sil+Vit E) (received SC and vitamin E) and SC+tramadol+vitamin E-treated group (Sil+Tra+Vit E) (received SC, tramadol and vitamin E). Drugs were administered orally once daily for four weeks after which blood samples were obtained and used to measure serum concentrations of liver enzymes (AST, ALT and ALP) and bilirubin. Results: Serum AST, ALT, ALP, total bilirubin (TB) and unconjugated bilirubin (UCB) concentrations were significantly (p<0.001) increased in Sil and Sil+Tra compared with control and Vit E and significantly (p<0.01) decreased in Sil+Vit E and Sil+Tra+Vit E compared with Sil and Sil+Tra. Serum conjugated bilirubin (CB) concentration and percentage conjugation of bilirubin significantly decreased (p<0.001) in Sil and Sil+Tra compared with control and Vit E and increased (p<0.01) in Sil+Vit E and Sil+Tra+Vit E compared with Sil and Sil+Tra. Conclusion: SC, administered separately and in combination with tramadol altered liver function. However, vitamin E supplementation was able to ameliorate this alteration in liver function.
