The COVID-19 pandemic demands changes in using of medicinal plants. Empirically, the medicinal plants of Curcuma xanthorrhiza (CX) and black cumin (BC) have been used massively in the era of the COVID-19 pandemic. Curcuma xanthorrhiza and black cumin (CXBC) preparations have been developed with the main ingredients of BC oil and CX extract. The purpose of the study was to determine the antioxidant and immune-modulatory activities of CXBC preparations active substances (polyphenols, flavonoids, thymoquinone). We conducted experimental laboratory research. The immunomodulatory activity test was carried out on human large lung cancer cell line (HTB-183 cells) by observing the expression of tumor necrosis factor alfa (TNF-α) and interleukine 10 (IL-10). The results showed that the CXBC preparation contained 4% thymoquinone, 25.87 mg/ml polyphenols, and 41.86 mg/dl flavonoids. CXBC preparations contain vitamins (A, C, and E) and minerals (potassium, calcium). The antioxidant activity of the CXBC preparation was included in the strong category with IC50=54.87 ppm. CXBC preparations increased TNF-a expression and decreased IL-10 expression in HTB-183 cells. Based on the study results, it can be concluded that the CXBC preparation contains 4% thymoquinone, 25.87 mg/ml polyphenol, 41.86 mg/dl flavonoid, and a high level of vitamin and minerals. CXBC preparations have potent antioxidant activity, increase TNF-α and decrease IL-10 expression.

Although several treatments are available for the treatment of type 2 diabetes mellitus, adverse effects and cost burden impose the search for safe, efficient, and cost-effective alternative herbal remedies. Syzygium aqueum (Burm.f.) Alston, a natural anti-inflammatory, antioxidant herb, may suppress diabetes-associated inflammation and pancreatic beta-cell death. Here, we tested the ability of the bioactive leaf extract (SA) to prevent streptozotocin (STZ)-induced oxidative stress and inflammation in pancreatic beta cells in rats and the involvement of the TLR-4 signaling pathway. Non-fasted rats pretreated with 100 or 200 mg kg−1 SA 2 days prior to the STZ challenge and for 14 days later had up to 52 and 39% reduction in the glucose levels, respectively, while glibenclamide, the reference standard drug (0.5 mg kg−1), results in 70% reduction. Treatment with SA extract was accompanied by increased insulin secretion, restoration of Langerhans islets morphology, and decreased collagen deposition as demonstrated from ELISA measurement, H and E, and Mallory staining. Both glibenclamide and SA extract significantly decreased levels of TLR-4, MYD88, pro-inflammatory cytokines TNF-α, and TRAF-6 in pancreatic tissue homogenates, which correlated well with minimal pancreatic inflammatory cell infiltration. Pre-treatment with SA or glibenclamide decreased malondialdehyde, a sensitive biomarker of ROS-induced lipid peroxidation, and restored depleted reduced glutathione in the pancreas. Altogether, these data indicate that S. aqueum is effective in improving STZ-induced pancreatic damage, which could be beneficial in treating type 2 diabetes mellitus.

Antioxidants have drawn the attention of the scientific community due to being related to the prevention of various degenerative diseases. The antioxidant capacity has been extensively studied in vitro, and different methods have been used to assess its activity. However, the main issues related to studying natural antioxidants are evaluating whether these antioxidants demonstrate a key role in the biological system and assessing their bioavailability in the organism. The majority of outcomes in the literature are controversial due to a lack of method standardization and their proper application. Therefore, this study aims to compile the main issues concerning the natural antioxidant field of study, comparing the most common in vitro methods to evaluate the antioxidant activity of natural compounds, demonstrating the antioxidant activity in biological systems and the role of the main antioxidant enzymes of redox cellular signaling and explaining how the bioavailability of bioactive compounds is evaluated in animal models and human clinical trials.
