Trabecular thickness measurement in cancellous bones: postmortem rat studies with the zoom-in micro-tomography technique

Abstract: Using the cross-sectional images taken with the zoom-in micro-tomography technique, we measured trabecular thicknesses of femur bones in postmortem rats. Since the zoom-in micro-tomography technique is capable of high resolution imaging of a small local region inside a large subject, we were able to measure the trabecular thickness without extracting bone samples from the rats. For the zoom-in micro-tomography, we used a micro-tomography system consisting of a micro-focus x-ray source, a 1248 x 1248 flat-panel… Show more

“…A calibration curve was constructed using six different concentrations (1,5,10,25,50, and 100 M) of Pro, Hyp, and PHP, and excellent linearity was obtained. The equations and correlation coefficients (r 2 ) of the calibration curves were as follows: Pro, y = 0.0294x + 0.0033, r 2 = 0.9999; Hyp, y = 0.0418x + 0.1071, r 2 = 0.9991; and PHP, y = 0.0113x + 0.0185, r 2 = 0.9990.…”

“…The induction of osteoporosis was confirmed 22 weeks after ovariectomy based on two-dimensional imaging of the trabecular bone of the left femoral neck. The images were obtained using micro-computed tomography (Micro-CT) according to a previously published method [10]. Mean trabecular thickness was used as the bone morphometric parameter.…”

“…Three-dimensional tomographic images were taken by micro-CT according to the previously published method. 32 Rats were anesthetized (1.5% isofluorane, 70% N 2 O, and 30% O 2 gas) and maintained on anesthetic gases for the duration of each measurement. The 3D images (512 · 512 · 512) were reconstructed from 450 projection data and the isotropic pixel resolution was 25 lm.…”

“…Particularly, trabecular bone imaging with a micro-CT is believed to be very useful for osteoporosis studies [5][6][7][8][9][10]. Longitudinal imaging of trabecular bone structure for a long time period through the life span of the animal subjects is often desired for evaluating the efficacy of new drugs or new therapeutic methods [11].…”
