TPA‐enhanced invasion of matrigel associated with augmentation of cell motility but not metalloproteinase activity in a highly metastatic variant (L‐10) of human rectal adenocarcinoma cell line RCM‐1

Abstract: We previously found that the enhanced activity to invade Matrigel upon stimulation with 12-O-tetradecanoylphorbol-13-acetate (TPA) was one of the major properties of a highly metastatic variant (L-10) of a human rectal adenocarcinoma cell line RCM-1. To clarify the mechanism of this enhancement, we examined the effect of TPA on 2 major biological factors involved in tumor cell invasion: cell motility and matrix-degrading metalloproteinase activity. The enhanced invasiveness was inhibited by protein-kinase-C in… Show more

“…In this regard, the metastatic potential of B16 melanoma cells has been correlated with PKC activity and can be considerably enhanced with PKC activators [55]. Similar results have been reported for Lewis lung carcinoma cells [56], mouse mammary adenocarcinoma cell line [57], and more recently, for human colorectal [58], bladder [37], and gastric [59] carcinomas. However, this report is the first to suggest that there is a direct relationship between PKC and invasion in gliomas.…”

“…The cells showed a peak motile response at 20 ng/ml of TPA [8]. The migration was time dependent up to 24 h and approximately 80% of the maximum migration occurred by 6 h after TPA treatment [9].…”

“…The motility assay was run as described previously [8] with some modifications. Briefly, L-10 cells were seeded into compartments of an eight-well Lab-Tek tissue culture chamber slide (1.5 x 105 cells in 0.4 ml growth medium) and allowed to attach for 24 h at standard culture conditions (37°C, 5% CO2 in air, 100% humidity).…”
