Background: Schizophrenia is a serious psychiatric disorder that significantly affects the quality of life of patients. The objective of this study is to discover a novel antipsychotic candidate with highly antagonistic activity against both serotonin and dopamine receptors, demonstrating robust efficacy in animal models of positive, negative, and cognitive symptoms of schizophrenia.Methods: In the present study, we examined the activity of antipsychotic drug (NH300094) on 5-HT2A, 5-HT2C, 5-HT1A, 5-HT1B, 5-HT7, H1, M1, Alpha1A, D2L, D2S, Alpha2A, D3 receptor functional assay in vitro. In addition, multiple animal models, including dizocilpine (MK-801) induced hyper-locomotion; APO induced climbing; Conditioned Avoidance Response (CAR); DOI-Induced Head Twitch; Forced swimming test; Scopolamine induced cognitive impairment model, were used to verify the antipsychotic activity of NH300094 in preclinical.Results:In vitro functional assays have indicated that NH300094 is a potent antagonist of 5-HT receptors and dopamine receptors, with higher relative antagonistic activity against 5-HT2A receptor (5-HT2A IC50 = 0.47 nM) than dopamine receptors (D2L IC50 = 1.04 nM; D2S IC50 = 11.71 nM; D3 IC50 = 31.55 nM). Preclinical in vivo pharmacological study results showed that NH300094 was effective in multiple models, which is more extensive than the clinic drug Risperidone. Furthermore, the safety window for extrapyramidal side effects of NH300094 is significantly wider than that of Risperidone (For NH300094, mice catalepsy model ED50/ Mice MK-801 model ED50 = 104.6-fold; for Risperidone, mice catalepsy model ED50/ Mice MK-801 model ED50 = 12.9-fold), which suggests a potentially better clinical safety profile for NH300094.Conclusion: NH300094 is a novel potent serotonin and dopamine receptors modulator, which has good safety profile and therapeutic potential for the treatment of schizophrenia with cognition disorders.

Background: Patients with HIV-AIDS are at increased risk for both opportunistic infections, such as cerebral toxoplasmosis, and cognitive impairment. IgG anti-Toxoplasmosis gondii (T.gondii) is a marker for latent infection. However, its role in cognition in HIV patients remains unclear. The study aims to determine the correlation between Ig-G anti-T.gondii and each domain cognitive function in HIV patients with cerebral toxoplasmosis. Method: This was a cross-sectional study involving 110 HIV patients with cerebral toxoplasmosis who met the inclusion and exclusion criteria. We assessed cognitive function using Montreal Cognitive Assessment Indonesian Version (MoCA-INA) and measured the IgG anti-T.Gondii using the ELISA method. The Spearman correlation test was used to determine the correlation between Ig-G anti-T.gondii with each cognitive domain. Results: There was a significant correlation between IgG anti-T.gondii and cognitive function. (p = 0.004, r = -0.275). There was also significant relation between IgG anti-T.gondii with attention (p = 0.046, r =-0.19), abstraction (p = 0.036, r = -0.2), and delayed recall (p = 0.047, r =-0.19). But there was no significant relation between Ig-G anti-T.gondii with visuospatial (p = 0.171), naming (p = 0.521), language (p = 0.810), and orientation (p = 0.11) Conclusion: Lower Level Of IgG Anti-Toxoplasmosis Gondii Antibodies Is Associated With Worse Cognitive Function In Human Immunodeficiency Virus-Acquired Immunodeficiency Syndrome (HIV-AIDS) Patients with Cerebral Toxoplasmosis

El Toxoplasma gondii es un parásito que se encuentra, aproximadamente, en el 30 % de la población humana. Durante los últimos años se ha evidenciado que la infección latente puede ser un factor de riesgo para el desarrollo de trastornos mentales; particularmente para la esquizofrenia, ansiedad, trastornos bipolares y trastornos de conducta. La asociación con los trastornos neuropsiquiátricos pueden explicarse por la influencia que tiene el parásito sobre la expresión de múltiples neurotransmisores; entre ellos la dopamina. Se realizó una búsqueda en las bases de datos PubMed y SciELO de 2015 a 2023, se seleccionaron artículos originales y de revisión de revistas científicas internacionales, en idiomas inglés y español con el objetivo de describir la relación entre la seroprevalencia de T. gondii y el desarrollo de trastornos mentales en población adulta. Existe relación entre los trastornos mentales en la población adulta con la infección por Toxoplasma gondii y este aumenta la posibilidad de desarrollar esquizofrenia y depresión en individuos sin historial previo, y que podría exacerbar cuadros psiquiátricos previos con dificultad en el tratamiento. Sin embargo, no todos los datos estadísticos establecen una relación directa, algunos estudios demuestran una asociación, ciertos datos son discordantes, lo que abre una puerta para futuras investigaciones.
