Toxoplasma gondii serine-protease inhibitor-1: A new adjuvant candidate for asthma therapy

Abstract: Serine-proteases are important players in the pathogenesis of asthma, promoting inflammation and tissue remodeling. It’s also known that many serine protease inhibitors display immunomodulatory properties. TgPI-1 is a Toxoplasma gondii protein that exhibits broad spectrum inhibitory activity against serine proteases. In view of the increased prevalence of atopic disorders and the need to develop new treatment strategies we sought to investigate the potential of TgPI-1 for treating respiratory allergies. For th… Show more

“…In our study, HSP60 was identified just after the cat was infected with T. gondii , which further shows that HSP60 does play an important role in the differentiation of oocysts after cat infection. Serine protease inhibitors display immunomodulatory properties [ 46 ], and T. gondii serine protease inhibitor 1 (TgPI1) not only affects the virulence of T. gondii but also affects the differentiation of bradyzoites [ 47 ]. It has been speculated that this may occur through inhibition of T. gondii and/or host serine protease [ 47 ].…”

“…The full-length recombinant protein rTgPI-1 and its truncated versions, Nt (containing domains 1 and 2) or Ct (containing domains 3 and 4), were previously generated by Pszenny et al [12]. rTgPI-1 was expressed and purified as described by Soto et al [6]. Expression and purification of truncated proteins were performed using the same protocol as rTgPI-1.…”

“…Asthmatic mice intranasally treated with a mixture of rTgPI-1 and allergen showed higher reduction in asthma hallmarks compared with the administration of rTgPI-1 alone. Moreover, a reduction in allergen-specific T-cell proliferation in thoracic lymph nodes and spleen and an expansion of Tregs was only seen in the co-administered group, suggesting that this inhibitor may function as a tolerogenic adjuvant [6]. Hence, rTgPI-1 emerges as a novel adjuvant candidate to be included in SIT formulations.…”
