Toxicity screenings of nanomaterials: challenges due to interference with assay processes and components of classicin vitrotests

Abstract: Given the multiplicity of nanoparticles (NPs), there is a requirement to develop screening strategies to evaluate their toxicity. Within the EU-funded FP7 NanoTEST project, a panel of medically relevant NPs has been used to develop alternative testing strategies of NPs used in medical diagnostics. As conventional toxicity tests cannot necessarily be directly applied to NPs in the same manner as for soluble chemicals and drugs, we determined the extent of interference of NPs with each assay process and componen… Show more

“…However, these issues do not preclude the use of HTS approaches to screen for NM‐induced cytotoxicity 60. Assay interferences can be avoided by using label‐free methods 61, 62. Impedance‐based spectroscopy is suggested as a method that does not need markers or dyes, and that has enhanced sensitivity compared to traditional assays 63, 64.…”

“…3,[19][20][21][22] Although by this definition, a nanosheet with a 1 nm thickness is also considered as a NM, the term NM mostly refers to particulate structures consisting of solid materials for which the terms NMs and nanoparticles (NPs) are used interchangeably. The EMA defines nanotechnology as "the use of tiny structures -less than 1,000 nm across -that are designed to have specific properties".…”
