Toxicity, pharmacokinetics, and in vivo efficacy of biotinylated chitosan surface-modified PLGA nanoparticles for tumor therapy

Abstract: Based on our previous work on the PLGA nanoparticles modified with biotinylated chitosan (Bio-CS-PLGA NPs), we further studied the stability, toxicity, pharmacokinetics, and in vivo efficacy. The safety of NPs was studied through single-dose toxicity test in mice, and the result showed that NPs were well tolerated at the dose of 300 mg/kg. Compared with the free EPB group, the NPs group exhibited higher plasma drug concentration, longer half-life time. EPB-loaded NPs significantly inhibited the tumor growth co… Show more

“…Preparation of Bio-PA NPs EPI loaded Bio-PA NPs were prepared by the dialysis method according to our recently reported paper. 22,23 Briefly, 100 mg of Bio-PA and 5 mg EPI were dissolved in 4 ml dimethyl sulfoxide (DMSO). To form NPs, the solution was injected in the dialysis tubing to against 1000 ml deionized water and the dialyzed liquids were exchanged six times followed by sonication step using ultrasonic probe (Scientz JY92-II, Ningbo, China) at 100 W for 2 min.…”

“…Bio-PA NPs were prepared by the dialysis method according to our previous study. 22,23 TEM shows that both Bio-PA NPs and EPI-loaded NPs fabricated by dialysis method are spherical in shape (Figure 1). However, the mean diameter of the particle was increased from (130.9 ± 10.36) nm (Bio-PA NPs) to (169.6 ± 7.94) nm (EPI-loaded NPs) with the polydipersity index lower than 0.5, suggesting mono-dispersion, determined by dynamic light scattering.…”

“…Polymeric nanoparticles have been shown to be promising carriers for CNS drug delivery owing to their potential both in encapsulating drugs, hence protecting them from excretion and metabolism and in delivering active agents across the BBB without interfering with the normal function of the brain [41][42][43][44]. Recent studies have suggested that the size of the delivery vehicle is one of the pivotal aspects determining performance because it influences biodistribution, cellular uptake and circulatory half-life.…”

“…A hopefully progress has been reported for new treatments for diseases such as diabetes, spinal cord injury, Parkinson’s disease, and myocardial infarction by stem cells in USA (12). In addition, tumor therapy has been introduced in China and also in USA (31, 32). In Iran great advances in tissue engineering for vascular grafts, prognosis and treatment of breast cancer and acute kidney injury has been observed (33-35).…”
