Toxicity of two type II ribosome‐inactivating proteins (cinnamomin and ricin) to domestic silkworm larvae

Abstract: Cinnamomin and ricin are two type II ribosome-inactivating proteins. They exhibited a different toxicity to domestic silkworm (Bombyx mori) larvae by oral feeding bioassay. The LC50 of ricin to the silkworm larvae at third instar was much lower than that of cinnamomin. When the isolated 80S ribosome from domestic silkworm pupae was treated separately with the reduced cinnamomin or the reduced ricin, a specific RNA fragment (R-fragment) was produced as characterized by 8 M urea-denatured polyacrylamide gel (3.5… Show more

“…Several lines of evidence support the idea that ricin-B related lectins play a role in plant defense against pathogens (Chen et al, 2002b; Vandenbussche et al, 2004a,b) and insects (Wei et al, 2004; Shahidi-Noghabi et al, 2009). Over-expressing SNA-I′ or SNA-V from S. nigra in transgenic tobacco (Samsun NN) plants enhances the tobacco plant’s resistance against infection with tobacco mosaic virus.…”

“…Insecticidal activity of type-2 RIPs has been demonstrated towards some insects (Gatehouse et al 1990;Dowd et al 1998Dowd et al , 2003Wei et al 2004). Although the enzymatic mechanism of RIP activity is well defined, the physiological steps by which ribosome inactivation leads to cell death are not well understood.…”

“…Likewise, Bu et al (2008) showed that potato NAC domain proteins are induced rapidly and strongly after wounding but not systemically, and their transcript levels reach maximum after 1 h, consistent with our observations. Interestingly, JA is also reported to modulate expression patterns of ricin B lectins (type II ribosome inactivating proteins, RIPII) (Reinbothe et al 1994;Xu et al 2007) having negative impact on aphid growth (ShahidiNoghabi et al 2008(ShahidiNoghabi et al , 2009, shows toxicity to many other herbivores (Zhou et al 2000;Wei et al 2004;Van Damme 2008), and even reduces their rate of oviposition on seeds of chickpea (Sadeghi et al 2006). Our study shows that following 30 min of TMB feeding, RIPII transcript abundance increases more than fivefolds in the tolerant cultivar.…”
