Toxicity Evaluation and Biomarker Selection with Validated Reference Gene in Embryonic Zebrafish Exposed to Mitoxantrone

Abstract: Notwithstanding the widespread use and promising clinical value of chemotherapy, the pharmacokinetics, toxicology, and mechanism of mitoxantrone remains unclear. To promote the clinical value in the treatment of human diseases and the exploration of potential subtle effects of mitoxantrone, zebrafish embryos were employed to evaluate toxicity with validated reference genes based on independent stability evaluation programs. The most stable and recommended reference gene was gapdh, followed by tubα1b, for the 4… Show more

“…In order to reinforce the non-toxic effect of H. lanceolatum at the MNTC, we decided to further analyze its effects on key genes of hepatotoxicity ( fabp10a , gclc ), cardiotoxicity ( erg ) and nephrotoxicity ( ctgf ) ( Liu et al, 2018 ; Bauer et al, 2021 ) as well as fkbp5 , a gene known to be up-regulated by glucocorticoids ( Binder, 2009 ). To this aim, zebrafish eleutheroembryos were incubated from 3- to 5-dpf and processed for RT-qPCR analyses.…”

“…So mitoxantrone’s effects are likely attributed to multiple targets that are vital to cell survival [ 51 , 53 ]. Mitoxantrone treated cells have also shown some changes to gene expression profiles [ 57 , 58 ]. Our western analysis before and after mitoxantrone treatment show that in PEO1 cells mitoxantrone reduces RAD52 protein levels significantly, there is a significant increase in DNA damage and this synthetic lethality is part of the mechanism for the low EC 50 of 0.1 μM in PEO1 cells ( Fig 6E and 6F ).…”

“…Gclc is highly expressed in the zebrafish eye throughout embryonic development [29], and is involved in the zebrafish antioxidant response to xenobiotic exposure [8991]. Herein, we present a novel role for gclc in regulating zebrafish eye development; a reduction in gclc expression causes reduced eye area (microphthalmia).…”
