The increased use of nonsteroidal anti-inflammatory drugs (NSAIDs) has resulted in their ubiquitous presence in the environment. The toxicological properties of these 2 widely prescribed NSAIDs, namely racemic ketoprofen and its enantiomer S(+)-ketoprofen (dexketoprofen), were evaluated, firstly, by acute and chronic toxicity tests using 3 representative model organisms (Vibrio fischeri, Pseudokirchneriella subcapitata, and Ceriodaphnia dubia) and, secondly, by evaluating the responses of biotransformation systems and multidrug resistance-associated proteins (MRP1/MRP2) using the Poeciliopsis lucida hepatocellular carcinoma 1 (PLHC-1) fish hepatic cell line. Toxicity data from both acute and chronic dexketoprofen exposure indicated higher sensitivity through inhibition of bioluminescence and algal growth and through increased mortality/immobilization compared to racemic ketoprofen exposure. The growth inhibition test showed that racemic ketoprofen and dexketoprofen exhibited different effect concentration values (240.2 and 65.6 μg/L, respectively). Furthermore, racemic ketoprofen and dexketoprofen did not exert cytotoxic effects in PLHC-1 cells and produced compound-, time-, and concentration-specific differential effects on cytochrome P450 1A (CYP1A) and glutathione S-transferase levels. For CYP1A, the effects of racemic ketoprofen and dexketoprofen differed at the transcriptional and catalytic levels. Exposure to racemic ketoprofen and dexketoprofen modulated MRP1 and MRP2 mRNA levels, and these effects were also dependent on compound, exposure time, and concentration of the individual drug. The present study revealed for the first time the interactions between these NSAIDs and key detoxification systems and different sensitivity to the racemic mixture compared to its enantiomer. Sulfamethoxazole (SMX) is an old sulfonamide antibiotic that was launched first in combination with trimethoprim in 1969 by F.Hoffmann-La Roche. Although sales figures for SMX have been declining over the past 20 yr, the compound is still widely used; moreover, many measured environmental concentrations (MECs) are available from Europe, the United States, Asia, Australia, and Africa. To assess aquatic risks of SMX in Europe, the exposure of European surface waters was predicted based on actual sales figures from IMS Health, incorporating environmental fate data on one side, and based on collated MECs representing more than 5500 single measurements in Europe on the other. Environmental effects were assessed using chronic and subchronic ecotoxicity data for 16 groups of aquatic organisms, from periphyton communities to cyanobacteria, algae, higher plants, various invertebrates, and vertebrates. Predicted no-effect concentrations (PNECs) were derived using both deterministic and probabilistic methodology.

Pharmaceutical compounds represent a diverse set of biologically active molecules used in human and veterinary medicine. The increase in the consumption of these drugs contributes to the constant occurrence of these compounds in aquatic environments.The situation is even more serious in conditions of absence of sewage treatment system.This is the case of the city of Governador Valadares, located in the state of Minas Gerais, thus discharging all its effluent to the Doce river that supplies the region. In order to guarantee the quality of water, especially for human consumption, investments in additional treatments are necessary. Among these technologies, the use of membranes, which combine the stability of the process with the excellent quality of the treatment, stands out. Thus, nanofiltration (NF) and reverse osmosis (RO) membranes were tested aiming at the pharmaceutical compounds retention in the Doce river raw water. This study was divided into three steps. In the first one, pharmaceutical compounds occurrence in all samples of the Doce river raw water was investigated. Among the pharmaceutical compounds that were found, the following stand out: Phenylbutazone and Prednisone that were rejected in 98 and 80%, respectively, by the DK membrane, whereas Betamethasone was 40.85 removed by this membrane. In addition, the efficiency of Governador Valadares central water treatment plantwas also assessed, which showed a reduction in the concentration of the pharmaceutical products in the potable water when compared to the Doce river raw water. The second part investigated five NF and two RO membranes that were tested to evaluate the physico-chemical quality and the presence of pharmaceutical compounds in permeates. Additionally, the resistance, permeability and specific energy consumption (SEC) for each membrane were assessed. It was observed a removal high efficiency of the parameters evaluated for the RO membranes, however, these membranes presented the highest resistance, energy demandand lower permeate fluxes. Dense NF membranes presented abetter set of results. The third phase refers to the choice of the best operational condition, evaluating the best transmembrane pressure and feed flow for permeates production with high quality in the physical and chemical parameters, as weel asthe drugs removal compareted with the Doce river raw water. The best operational condition obtained in the study was 10 bar of pressure and feed flow of 3.2 L/min which presented permeates flow at 45.99 L / m².h and a SEC of 0.44 KWh.m³ m².
