Towards the elaboration of new gold-based optical theranostics

Abstract: Four new red BODIPY-gold(I) theranostic compounds were synthesized. Some of them were vectorized by tethering a biovector (glucose or bombesin derivatives) to the metallic center. Their photophysical properties were studied. Additionally, their cytotoxicity was examined on different cancer cell lines and on a normal cell line, they were tracked in vitro by fluorescence detection, and their uptake was evaluated by ICP-MS measurements.

“…Another explanation could be that the proximity of the coumarin to the imidazole ring may cause a photoinduced electron‐transfer phenomenon, but ligand 1 displays an interesting quantum yield. Otherwise, it is worth noting that, as observed in previous studies, coordination to the gold(I) ion does not quench the fluorescence, but even increases the quantum yields of 1 and 2 by a factor of two.…”

“…Numerous strategies explored the attachment of a targeting ligand (i.e. sugars and peptides) through S-Au bonds (Bertrand et al, 2014;Doulain et al, 2015). Although some drawbacks, particular promising are those exploiting these bonds for the functionalization with molecules for biosensing, anticancer drug delivery (i.e.…”

“…In addition, a number of strategies explored the attachment of a targeting ligand (i.e., sugars and peptides) through S−Au I bonds. However, functionalisation of the complex with a directing ligand has often led to a decrease in activity . A number of reviews summarizing the role of NHC−Au and S−Au conjugates in biomedicine, in particular as anticancer agents, have been published …”
