Towards multimodal nonlinear optical tomography - experimental methodology

Abstract: All-optical microspectroscopic and tomographic tools reveal great potential for clinical dermatologic diagnostics, i.e., investigation of human skin and skin diseases. While optical-coherence tomography has been complemented by twophoton fluorescence tomography and second-harmonic generation tomography, a joint study of various nonlinear optical microspectroscopies, i.e., application of the recently developed multimodal imaging approach, to sizable human-tissue samples has not been evaluated up to now. Here, w… Show more

“…5 where CARS and SHG have been measured simultaneously on cardiomyocyte cells. CARS has also been used along with TPAF for skin imaging 137,138 or small organism imaging. 139 As several excitation sources are required for CARS, other nonlinear frequencies such as SFG can be implemented directly, and possibly simultaneously.…”

“…The SHG images used both for training and validation of the herein presented algorithm were recorded as an integral part of the multimodal imaging approach described in detail by Vogler and coworkers [38]. In contrast to CARS highlighting the spatial distribution of lipids within the sections [39,40] and TPF, which displays the presence of autofluorophores within the sample, the SHG images used here highlight highly ordered structures lacking inversion symmetry in the sample.…”
