Total polyphenol of Anemarrhena asphodeloides ameliorates advanced glycation end products‐induced endothelial dysfunction by regulation of AMPKinase (知母多酚通过激活AMP‐激酶来缓解糖基化终末产物引起的内皮损伤)

Abstract: Total polyphenol of Anemarrhena asphodeloides inhibited AGEs-induced ROS-associated inflammation and ameliorated endothelial dysfunction through beneficial regulation of AMPK activation.

“…EXO-FBS500A-1; System Biosciences, Palo Alto, CA, USA). According to our previous study (21), the PVAT samples were cultured with PA (100 µM) in the presence or absence of mangiferin (0.1, 1 or 10 µM) for 2 h; samples cultured without PA or mangiferin were used as the control group. Subsequent to washing with PBS, PVAT samples were incubated in DMEM for a further 22 h. The supernatant was then collected as the conditioned medium (CM), and larger particles were removed by centrifugation at 10,000 × g for 5 min at 4°C and filtration through a 0.22-µm filter.…”

“…Mangiferin regulates AMP-Kinase (AMPK) activity and this action is involved in amelioration of metabolic disorders [19,20]. Our previous works also showed that mangiferincontaining total polyphenol of Anemarrhena asphodeloides ameliorated endothelial dysfunction with regulation of AMPK activity [21]. AMPK is an energy sensor with antiinflammatory activity.…”

“…The rhizomes of A. asphodeloides , known as Zhimu ( Anemarrhenae Rhizoma ), have been widely used in the treatment of febrile diseases, hyperpyrexia, polydipsia, cough, osteopyrexia, fever and constipation since ancient times (Chinese Pharmacopoeia, ). Modern pharmacological studies have shown that A. asphodeloides has obvious effects including anti‐tumor (Guo et al, ), anti‐oxidation (Chae et al, ), anti‐inflammation (Bak et al, ) and anti‐diabetes (Zhao et al, ). Meanwhile, it has exhibited neuroprotective activities for central nervous system disorders such as Alzheimer's and Parkinson's disease (Lee, Jung, & Kim, ; Won & Ma, ).…”
