“…The combination of TLI and low-dose cyclosporine was found to be effective and clinically safe in rats, 216 and TLI with postoperative antithymocyte globulin induced permanent and specific transplantation tolerance toward heart allografts in about 40% of transplanted dogs. 249 These encouraging results led to a similar trial in clinical kidney transplantation (discussed later).…”

“…The effect of TLI is dose dependent, however, several aspects of how treatment is given seem to 14 be essential (143,144) and have been the subject to some controversary (145). Preoperative timing, the dose given per session and dose rates may be critical for an optimal recipient preconditioning.…”

“…Cyclosporin (CyA) has obvious appeal for use after TLI in view of its marrow-sparing properties. Rynasiewicz et al [26] found a marked synergistic effect between 1,000 cGy of TLI and low doses of CyA (1.25 or 2.5 mg/kg per day) in heterotopic heart transplantation in rabbits. On the other hand, the addition of CyA to TLI in heart transplantation in cynomolgus monkeys did not result in a significant increase in graft survival [24].…”
