Total Flavonoids of Chuju Decrease Oxidative Stress and Cell Apoptosis in Ischemic Stroke Rats: Network and Experimental Analyses

Abstract: Background: Pharmacological research results showed that total flavonoids of Chuju (TFCJ) could be used to treat acute myocardial ischemia and myocardial ischemia-reperfusion injury. In this study, we explored the protective effect of TFCJ on ischemic stroke (IS) in the IS rat model. We hypothesized that TFCJ might exert its neuroprotective effects by suppressing apoptosis and oxidative stress that are closely related to PI3K/Akt/mTOR signaling pathway.Method: TFCJ (10, 20, and 40 mg/kg) was administered for 7… Show more

“…In addition, flavonoids can exert an effect on the expression, as well as on the activity of apoptotic proteins such as BCL-2, BAX, and cleaved caspase-3 (CASP3), which ultimately leads to the induction of apoptosis in cancer cells. Polyphenols are found in a variety of plant-based foods and beverages [ 33 , 35 ]. They can influence the process of PI3K/AKT/mTOR signaling, which is necessary for the survival of cells, the proliferation of cells, the metabolism of cells, and angiogenesis.…”

“…It catalyzes the iterative condensation and intramolecular cyclization of one molecule of p -coumaroyl-CoA and three molecules of malonyl-CoA to produce naringenin chalcone, which is the basic flavonoid skeleton . Molecular modification by downstream isomerases, hydroxylases, reductases, glycosyltransferases, and acyltransferases biosynthesizes a wide range of flavonoids from naringenin chalcone. , Naringenin chalcone and its derived flavonoids are widely distributed in plants and constitute a major class of natural products, including more than 15000 compounds. , Flavonoids have numerous applications in medicine, agrochemicals, cosmetics, food, and the chemical industry because of their various physiological activities, such as antiatherosclerotic, oxidation resistance, liver protection, and anticarcinogenic. , CHS has become a focus of research attention because of its essential function in the flavonoid biosynthetic pathway …”

“…Proteins, DNA, and fats in cells undergo crucially significant modifications as a consequence of excessive ROS production [13]. Several investigations have demonstrated a strong association between OS and neurological conditions such as stroke, AD, and TBI [14–17]. An essential transcription agent called nuclear factor erythroid 2 related factor 2 (Nrf2) is crucial in the fight against OS.…”

“…Along with cherries and other berries, apples have the greatest concentration of Que among fruits [33, 34]. Que is a healthy dietary component with few side effects [15]. The elimination of ROS is induced by the Que therapy, and brain tissues are protected from OS, according to other reports [35, 36].…”
