“…On the one hand, this indicates that at least in some of the animals, circulating antibodies might have found virulent virus administered by the oronasal route. On the other hand, these observations are concurrent with previous findings by our group and others regarding the potential in vivo FMDV-neutralizing roles for the bovine IgM in naive cattle at early times postinfection (22) and the IgG1 isotype in vaccinated animals at later times postimmunization (24,28). Most significantly, we described for the first time that, besides the systemic and ASC responses in the draining lymph node, intramuscular (systemic) FMD vaccination in the neck also was capable of promoting local virus-specific ASC in respiratory lymphoid tissues in cattle.…”

“…FMDV-specific isotype ELISA was performed as reported by Capozzo et al (30), except that sheep anti-bovine IgG1 and IgG2 HRP-conjugated antibodies were used at a dilution of 1:750, and sheep anti-bovine IgM HRP-conjugated antibody was used at 1:5,000 (BD-Serotec, Oxford, United Kingdom).…”

“…The response is directed to epitopes on the three external structural proteins (see "Antigenic variation" above), and good protective immunity is apparent between 7 and 14 days after either infection or vaccination. In cattle, the immunoglobulin G1 (IgG1) response predominates over IgG2 (86,326,417), and antibody, including IgA, can be detected in upper respiratory secretions early in infection (415,417). The neutralization of virus within the host may occur by mechanisms similar to those occurring in in vitro neutralization; however, there is a suggestion that macrophages may play a role in clearing the virus from the infected animal by phagocytosis of opsonized virus (306,307,383).…”
