TorsinA Is Functionally Associated with Spermatogenesis

Abstract: TorsinA is a member of the AAA+ superfamily of adenosine triphosphatases. These AAA+ proteins have numerous biological functions, including vesicle fusion, cytoskeleton dynamics, intracellular trafficking, protein folding, and degradation as well as organelle biogenesis. Of particular interest is torsinA, which is mainly located in the endoplasmic reticulum (ER) and nuclear envelope (NE). Interestingly, mutations in the TOR1A gene (the gene encoding torsinA) are associated with DYT1 dystonia and with the prefe… Show more

“…The top ranking of TOR1A in the normal prostatic tissue justifies its major roles in protein folding, degradation and trafficking, vesicle fusion, cytoskeleton dynamics, organelle biogenesis, and spermatogenesis [ 70 ]. We found TOR1A as significantly up-regulated in the nodule “B” ( x = 1.44), but not altered in the other two cancer nodules.…”

“…Interestingly, the first two genes for P1 are: The uncharacterized LOC145474 and the pseudogene SCARNA7 (also known as U90 ), the latter being associated with the non-small cell lung cancer[ 83 ]. The top 2 genes in P2C are: DENND1B , associated with gastric cancer[ 84 ] and TOR1A associated with dystonia and spermatogenesis[ 85 ]. Note the lack of overlap between the top 10 genes of the two persons and that the GCHs of the cancer top genes have much lower scores in the normal tissue.…”

“…TOR4A, TorsinA, is a member of the AAA + ATPase family that is located within the lumen of the endoplasmic reticulum and nuclear envelope. AAA + proteins are associated to several biological functions, such as vesicle fusion, cytoskeleton dynamics, intracellular trafficking, and protein folding (Serrano et al, 2019). TorsinA is linked to regulation of protein trafficking quality control and nuclear envelope function (Gonzalez-Alegre and Paulson, 2004;Nery et al, 2011).…”
