Topology of phosphatidylserine synthase 1 in the endoplasmic reticulum membrane

Abstract: Phosphatidylserine (PS) synthase 1 (PSS1) of mammalian cells is a multiple membrane‐spanning protein of the endoplasmic reticulum (ER) and regulated by inhibition with the product PS. Alanine‐scanning mutagenesis of PSS1 has revealed eight amino acid residues as those crucial for its activity and six as those important for its regulation. Furthermore, three missense mutations in the human PSS1 gene, which lead to regulatory dysfunctions of PSS1 and are causative of Lenz–Majewski syndrome, have been identified.… Show more

“…PSS1 topology and locations of previously reported missense pathogenic variants in PSS1. Topology is based on the 10 transmembrane‐helix model described by Miyata and Kuge (2021) and the boundaries of transmembranes designated by numbers is defined using the AF‐P48651‐F1‐model_v2. Pdb ( https://alphafold.ebi.ac.uk/entry/P48651).…”

“…PSS1 catalyzes the exchange of choline from phosphatidylcholine for serine, whereas PSS2 performs the equivalent exchange with ethanolamine from phosphatidylethanolamine. In both cases, phosphatidylserine is obtained [34,35].…”

“…Specifically, PSS1 exchanges the choline of a PC for a serine, generating a PS molecule. However, PSS2 changes the ethanolamine of PE for a serine, also generating PS [11,14,15].…”

“…However, there is also another pathway like PC synthesis, whereby ethanolamine is phosphorylated by ethanolamine kinase and then converted to CDP-ethanolamine. CDP-ethanolamine is finally converted to PE when DAG is incorporated [14,15]. Although PC, PE, PS, PI, CL, and PG are all derived from phosphatidic acid, they follow different routes.…”
