Topographic Restriction of TAG-1 Expression in the Developing Retinotectal Pathway and Target Dependent Reexpression during Axon Regeneration

Abstract: TAG-1, a glycosylphosphatidyl inositol (GPI)-anchoredprotein of the immunoglobulin (Ig) superfamily, exhibits an unusual spatiotemporal expression pattern in the fish visual pathway. Using in situ hybridization and new antibodies (Abs) against fish TAG-1 we show that TAG-1 mRNA and anti-TAG-1 staining is restricted to nasal retinal ganglion cells (RGCs) in 24-to 72-h-old zebrafish embryos and in the adult, continuously growing goldfish retina. Anti-TAG-1 Abs selectively label nasal RGC axons in the nerve, opti… Show more

“…Of these genes, gap43 (Reinhard et al, 1994) and tag1/axonin (cntn2 -Zebrafish Information Network) are expressed by migrating cerebellar granule cell precursors (Fig. 3E,G) (Console-Bram et al, 1996;Wolfer et al, 1994;Lang et al, 2001). In mutant embryos, no expression of these markers can be detected in the region that would give rise to the cerebellum, at any of the stages investigated (Fig.…”

“…Zebrafish embryos were processed for whole-mount immunohistochemistry as previously described (Marx et al, 2001;Weiland et al, 1997). Embryos were labeled with zebrafish pAb Tag-1 (axons, Lang et al, 2001), anti-␣-actinin (Biotrend), antiphospho-Tyrosin (myotome boundaries, Santa Cruz Biotechnologiy), and phalloidin-Alexa 488 (actin, Invitrogen). Embryos were analyzed using a confocal microscope (LSM 510, Zeiss) equipped with a high aperture lens 40ϫ/1.2W (C-Apochromat, Zeiss) and the appropriate lasers.…”

“…TAG-1 can bind homophilically or heterophilically with other members of the Ig superfamily to mediate adhesion or signaling between cells or axons (Furley et al, 1990;Kuhn et al, 1991;Rader et al, 1993;Felsenfeld et al, 1994;Fitzli et al, 2000;Perrin et al, 2001;Pavlou et al, 2002;Karagogeos, 2003). We examined TAG-1 protein distribution on R-B axons by labeling with an anti-TAG-1 antibody (Lang et al, 2001). TAG-1 protein was expressed strongly on the central R-B axons at all stages that were examined, 20 -27 hpf ( Fig.…”
