Aggregation of nicotinamide (3-pyridine-carboxamide; NA) and picolinamide (2-pyridinecarboxamide; PA) has been investigated by matrix-isolation, supersonic jet and neat solid state infrared spectroscopy, complemented by DFT(B3LYP)/6-311++G(d,p) calculations. For both compounds, the most stable dimeric structure was shown to be the centrosymmetric dimer where two monomers in their most stable forms establish two NHÁ Á ÁOQC hydrogen bonds. The most stable structures of monomers of NA and PA were characterized in detail experimentally by matrix-isolation spectroscopy and theoretically (at both the DFT(B3LYP)/6-311++G(d,p) and MP2/6-311++G(d,p) levels). For nicotinamide, two conformers were found in the matrices, with ca. 80% of the total population adopting the E form. The monomers and dimers of PA and NA were also investigated by infrared spectroscopy of the studied compounds seeded in supersonic jet expansions. These studies revealed that the constraints on the vibrational dynamics in the PA dimer are different from those in the NA dimer. In the PA dimer, the vibrational energy flow out of the N-H stretching mode was shown to be accelerated substantially by the presence of a secondary intramolecular hydrogen bond. In the glassy state of both compounds, the centrosymmetric dimer seems to be the prevalent structure. In the neat crystalline state (KBr pellet), picolinamide keeps this type of dimeric structure as the constituting unit, whereas nicotinamide molecules assume a different arrangement where one of the NHÁ Á ÁOQC bonds is replaced by an NHÁ Á ÁN(ring) bond. The different crystallograpic structures which were formed by the compounds are reflected in the vibrational spectra of the solids. These observations are correlated with the molecular properties of NA and PA, in particular with the greater conformational mobility of NA compared with PA. This is ascribable to the absence in the NA molecule of the intramolecular NHÁ Á ÁN (ring) interaction, which exists in PA.

Through its involvement in over 400 NAD(P)-dependent reactions, niacin status has the potential to influence every area of metabolism. Niacin deficiency has been linked to genomic instability largely through impaired function of the poly ADP-ribose polymerase (PARP) family of enzymes. In various models, niacin deficiency has been found to cause impaired cell cycle arrest and apoptosis, delayed DNA excision repair, accumulation of single and double strand breaks, chromosomal breakage, telomere erosion and cancer development. Rat models suggest that most aspects of genomic instability are minimized by the recommended levels of niacin found in AIN-93 formulations; however, some beneficial responses do occur in the range from adequate up to pharmacological niacin intakes. Mouse models show a wide range of protection against UV-induced skin cancer well into pharmacological levels of niacin intake. It is currently a challenge to compare animal and human data to estimate the role of niacin status in the risk of genomic instability in human populations. It seems fairly certain that some portion of even affluent populations will benefit from niacin supplementation, and some subpopulations are likely well below an optimal intake of this vitamin. With exposure to stressors, like chemotherapy or excess sunlight, suraphysiological doses of niacin may be beneficial.

ÖzetAmaç: Bu çal›flmada topikal niasinamidin hafif ve orta fliddetli akne vulgaris tedavisinde etkinli¤i ve güvenirli¤i ve yaflam kalitesi üzerine etkisi araflt›r›ld›. Gereç ve Yöntem: Hafif ve orta fliddetli derecede aknesi olan, dermatoloji poliklini¤ine baflvuran 29 kad›n hasta çal›flmaya al›nd›. Hastalar sekiz hafta boyunca yüzlerine günde iki kez %4 niasinamid içeren krem-jel (acnecinamide-Microgen) uygulad›lar. Lezyonlar›n say›s› (noninflamatuvar ve inflamatuvar) 0,2,4 ve 8. haftalarda say›ld›. Yan etkiler (eritem, deskuamasyon yanma, ve kserozis) kaydedildi. Hastalar tedavinin bafl›nda ve sonunda dermatolojiye özel yaflam kalite ölçe¤i olan Skindeks-29'u tamamlad›. Bulgular: Tedavi sonunda inflamatuvar lezyonlar›n say›s›nda azalma istatistiksel olarak anlaml›yd› (12,24 6,14, p=0,000). Fakat noninflamatuvar lezyonlarda tedavinin bafl›nda ve sonundaki azalma istatistiksel olarak anlaml› de¤ildi. Niasinamid krem-jel iyi tolere edildi. Skindeks-29 skala ortalamalar›nda istatistiksel olarak anlaml› düzelme sa¤land›.
