Topical Application of Cyclosporine on Guinea Pig Allergic Contact Dermatitis

Abstract: Topical cyclosporine applied to the test site substantially inhibited the elicitation reaction of contact sensitivity in the guinea pigs previously sensitized with 2,4-dinitrochlorobenzene (DNCB). This suppressive effect of the drug was short lived and reversible. Cyclosporine was not effective when given six hours or later after antigenic challenge to the test site. Cyclosporine had no effect on the toxic contact reaction in normal animals either to croton oil or to DNCB in high concentration. Cyclosporine ap… Show more

“…The acute hypersensitivity reaction was associated with a pronounced erythema and accompanying oedema of all the test sites and the reaction was fully developed 24 h after elicitation. This animal model has been used as a suitable model for ACD and appears to be an appropriate model to evaluate topical anti‐inflammatory compounds 16–21 …”

“…Contact dermatitis was induced, as described by several investigators 16–21 . Guinea pigs were shaved and four 2‐cm diameter areas were tattooed on their backs using a marker, two left and two right of the spinal midline.…”

“…All animals had the area behind both ears shaved, and 24 h later guinea pigs were sensitized with 5% DNCB solution (DNCB dissolved in olive oil: ethanol mix 1:4) behind one ear whereas mice were sensitized with 1% DNCB solution to an area behind one ear (17).…”

“…The contact hypersensitivity response (CHR) elicited by a single epicutaneous application of a contact sensitizing agent in a presensitized animal has been widely used as a test system to evaluate the effects of topical and systemic anti‐inflammatory compounds including corticosteroids and immunosuppressants 1–3 . These acute CHR models are simple, rapid and reproducible, and are thus suitable for preliminary screening purposes and for elucidating the mechanism of action of anti‐inflammatory compounds.…”
