Toll-like Receptors as a Target of Food-derived Anti-inflammatory Compounds

Abstract: Background: Vegetable-rich diets are associated with the reduced risk of inflammatory diseases. Results: Iberin, an isothiocyanate compound from cabbage, targeted TLRs, disrupted TLR dimerization, and inhibited the inflammatory responses. Conclusion: The TLR dimerization step is a target of food-derived anti-inflammatory compounds. Significance: These findings extend our understanding of how isothiocyanates show their medical benefits as anti-inflammatory food components.

“…Among 40 natural compounds docked to IRAK-4, apigenin, bilobetin, curcumin, ginkgetin, quercetin, berberine, and pseudopterosin showed higher affinity compared to the native ligand. Quercetin was reported to show IRAK-4 inhibitory activity [40]. Affinity of quercetin to the binding site was supported by hydrogen bond between 4-hydroxy of B-ring with Lys213B and Glu233B residues, while the aromatic rings contributed by forming hydrophobic interaction with Val200B, Tyr262B, and Leu318B.…”

“…Moreover, an anti-inflammatory effect mediated via attenuation of tumor necrosis factor α (TNF-α), NFκB and mitogen-activated protein kinases (MAPK), as well as depletion of IL-6, IL-1β, IL-8 or monocyte chemoattractant protein-1 (MCP-1) gene expression has also been attributed to this polyphenol [143]. …”
