Toll-Like Receptors and Agonist Responses in the Developing Fetal Sheep Lung

Abstract: Toll-like receptors (TLRs) are pattern recognition molecules that initiate innate immune responses. Intra-amniotic exposure of fetal sheep to pro-inflammatory stimuli causes pulmonary inflammation and induced lung maturation. We examined TLR ontogeny and fetal lung responsiveness to three different TLR agonists. We cloned ovine TLRs 2, 3, and 4 and found 83-88% homology between these ovine and human TLRs. Lung TLR2 and 4 mRNAs increased throughout late gestation to 50% of adult level in the term newborn lamb. … Show more

“…Bronchoalveolar lavage fluid (BAL) of the left lung was used to determine total protein content (23) and differential cell counts after cytospins. Tissue from the left lung was snap frozen for RNA isolation, and 10 g of total RNA was used for IL-1␤ and IL-6 RNAse protection assays (24,25). The right upper lobe was inflation fixed with 10% formalin (26), and this tissue was used for injury scores.…”

“…Protein (23), and cell counts and differentials using cytospins were measured from the BALF. Total RNA was isolated from lung and liver, and 10 g of total RNA was used for IL-1␤, IL-6, IL-8, and TLR-4 quantitation using RNAse protection assays (24,25). LPS was measured in plasma collected after 3 h of ventilation using a Limulus amebocyte lysate assay (Bio Whittaker, Walkersville, MD) (16).…”

“…70 The lung inflammation could be induced by LPS from periodontal organisms, 71 but a synthetic agonist for Toll-like receptor (TLR)-2 did not cause a consistent fetal lung inflammation. 72 Intraamniotic injections of endotoxin at 60 to 80 days of gestational age (40 to 55% of gestation) resulted in fetal lung inflammation at 125 days (term 150 days). 73 The experimental sheep work therefore suggests that the immature fetus can mount inflammation after exposure to some proinflammatory stimuli, with subsequent structural changes suggestive of lung maturation.…”
