Tolerogenicity of resting and activated B cells.

Abstract: SummaryAntigen presentation by resting splenic B cells has been shown previously to induce T helper I cell (Thl) anergy. In contrast to expectations, it was found here that B cells treated with F(ab')2 goat anti-mouse immunoglobulin (IgM) for 24 or 48 h also presented antigen (Ag) to Thl cells in a manner that induced dramatic Ag-specific proliferative inactivation. The tolerogenicity of the anti-Ig-treated B cells was consistent with the observation that these B cells were only slightly more efficient than re… Show more

Small B lymphocytes express only low levels of the B7-1/2 costimulatory molecules on ance using intrathymic peptides appeared to be mediated by the induction of T-cell anergy because the tolerant their surface; unless stimulated, they are ineffective as APC (116,158,296). Small B cells induce tolerance when acting state could be broken by administration of exogenous IL-2 (26, 235).…”

“…Recently, it was shown that on a per cell basis B lymphocytes are as effective as DC in activating naive T cells [4] and that B lymphocytes and DC undergo a similar increment of peptide/MHC Class II complexes after intravenous injection of antigen [5]. In spite of this evidence, several reports indicate that B lymphocytes fail to activate naive T cells [6,7].…”

“…Because resting B cells are known to express low levels of costimulatory molecules and can tolerize the T cells (5,11,12), we therefore used activated B cells as APC for our study. It is known that B cell activation may occur in response to two kinds of Ags, T-dependent and T-independent Ags (13).…”
