The study, expansion and preservation of the genetic diversity of the source material, and its purposeful use in hybridization is the basis for the creation of adaptive varieties of durum spring wheat that are resistant to biotic and abiotic factors of the environment of Western Siberia. The objects of research were samples of durum spring wheat. Over the years of research , about 3 thousand samples were worked out from the world gene pool of various countries and regions: from the collection of the VIR, the gene pool from Mexico (CIMMYT) within the framework of the agreement and cooperation program (2000)(2001)(2002)(2003)(2004)(2005)(2006)(2007), from 2000 to the present time under the program of the Kazakh-Siberian Spring Wheat Breeding Network (KASIB), from other scientif ic institutions of Russia in exchange activities. Using generally accepted techniques, the obtained material was studied for a complex of traits: yield, adaptability, grain quality, resistance to diseases. In the cycle of studying the gene pool from CIMMYT, 50 genotypes were identif ied in terms of yield at the level of the Omskaja jantarnaja standard, 276 grains by test weight, 131 samples by pasta color, 131 samples by resistance to hard smut, and 112 by resistance to powdery mildew. Almost all samples were not affected by leaf rust. The study set showed high sensitivity to extreme conditions and most forms of interest in quality and disease resistance were low-productive in our environment. In KASIB nurseries, 29 samples were identif ied in terms of yield and adaptability, 29 samples in terms of grain quality, 21 in terms of resistance to diseases, including 8 resistant to stem rust. In the set of varieties received from the VIR, 15 genotypes were adaptive, 16 had high grain quality, 11 were resistant to stem rust. In the breeding material, 17 samples of the local population resistant to stem rust (6 of them were comprehensively resistant) and 25 race-resistant to Ug99 were identif ied. The genotypes identif ied as a result of research are of interest as sources of valuable traits.

Kursk Federal Agricultural Research Center, together with National Grain Center named after P.P. Lukyanenko, has been studying and selecting hybrid populations, lines and varieties of hard winter wheat since 2012. Since 2017, they have been studied for ecological plasticity and the presence of economically valuable features when cultivating in the Central Chernozem Region (Kursk Region) varieties of winter hard wheat bred by FSBSI “Agricultural Research Center “Donskoy”on the basis of agreements on creative and scientific cooperation. At this stage of research, a large number of seedlings of winter hard wheat were tested, of which the most promising ones were identified. Using highly effective methods of ecological selection and having worked out breeding material in the environmental breeding laboratory of Kursk FARC, research data for 2018-2020. on yield, resistance to creep, overwintering, crop structure (height of plants, number of plants, number of stems, number of productive stems, total bush, productive bush, mass of 1000 grains, length of main spike, number of grains in main spike, weight of grain from one plant) and grain quality, on the basis of which grades and lines of winter hard wheat are evaluated. Over the years of research, weather conditions have developed in a variety of ways. Adverse environmental factors made it possible to better assess the level of adaptability of the studied sorto samples to abiostresses. In this regard an important task of joint breeding, is to increase environmental sustainability, the ability to ensure high stable yields in the soil and climatic conditions of the cultivation region.

In order to obtain high yields of winter durum wheat, it is necessary not only to use advanced agricultural technologies, but also to sow varieties resistant to major diseases. One of these diseases that cause great harm to grain crops is Septoria leaf blotch. The current paper has presented the study results of collection material of Russian and foreign winter durum wheat according to the efficiency of the Septoria leaf blotch resistance gene Stb2 in the North Caucasus region. The purpose of the current study was to identify the Septoria leaf blotch resistance gene Stb2 in the collection material of winter durum wheat. The study was carried out in 2017–2018 in the laboratory of marker breeding of the FSBSI “ARC “Donskoy” in Zernograd. In the course of the study there have been applied such methods as the isolation of genomic DNA by the CTAB-method, PCR-analysis with the molecular SSR marker WMS533, electrophoresis on 2% agarose gel, analysis of electrophoresis gels in the Bio-Rad Image Lab 6.0 software, general data analysis by Microsoft Excel. The study has shown that 29 samples out of 185 analyzed ones had the Septoria leaf blotch resistance gene Stb2, they are the lines L3557 h42, L3182 h7, L3335 h29, L3336 h43, L3408 h218 (Russia, FSBSI “Research Center of Plants named after P. P. Lukyanenko”), K-61620 (Russia), K-54455, DF 917 (Romania), K-61924, K-59179, K-51863, K-61869 (Moldova), K-63871 (Turkey), Kharkovskaya 909 (Ukraine), MVTD-15-99 (Hungary), 2196-1/04, 922/93, 589/13, 655/13, 683/13, 145/14, 322/14, 484/14, Gordeiform 1424/82 (Russia, FSBSI “ARC “Donskoy”), Beliy Parus (Ukraine), Belgorodskaya Yantarnaya (LLC “Sativa” in Belgorod, Russia), IR named after V. Ya. Yuriev (Kharkov, Ukraine)), Zhivitsa (Russia, Prikumskaya OSS), Pandur (Romania) and Elidur (Romania). There have been recommended using the identified samples of winter durum wheat with the Septoria leaf blotch resistance gene Stb2 in breeding programs.
