Toc12, a Novel Subunit of the Intermembrane Space Preprotein Translocon of Chloroplasts

Abstract: Translocation of proteins across membranes is essential for the biogenesis of each cell and is achieved by proteinaceous complexes. We analyzed the translocation complex of the intermembrane space from chloroplasts and identified a 12-kDa protein associated with the Toc machinery. Toc12 is an outer envelope protein exposing a soluble domain into the intermembrane space. Toc12 contains a J-domain and stimulates the ATPase activity of DnaK. The conformational stability and the ability to stimulate Hsp70 are depe… Show more

“…In other cases, the Hsp70 and type III J-domain protein are recruited independently to the site of action, where the J-domain protein stimulates ATP hydrolysis by the partner Hsp70. This can be observed for the import machinery of mitochondria and chloroplasts (Westermann and Neupert, 1997;Becker et al, 2004;Kozany et al, 2004;Qbadou et al, 2007). The divergence of the C terminus among the type III J-domain proteins and their uniqueness suggests the possibility that they bind very different proteins.…”

“…Tic21 was recently identified by Arabidopsis genetics (Teng et al, 2006). When antibodies against pea Tic110 and SPA-820 monoclonal antibody (StressGen), which recognizes an as yet unidentified intermembrane space Hsp70 (Schnell et al, 1994;Becker et al, 2004), were added, the mobility of the 1-MD complex on BN-PAGE was not affected ( Figure 5A). All other antibodies, which we tested, against known translocon components failed to shift the 1-MD complex by antibody-shift BN-PAGE (i.e., antibodies against Toc159, Toc34, Tic40, Tic22, Cpn60a, Cpn60b, and stromal Hsp70) (see Supplemental Figure 6 online).…”

“…The existence of Toc12 in IMS, however, has been controversially discussed in the recent years (Becker et al , 2004a ;Chen et al , 2010 ;Chiu et al , 2010 ). Toc12 was annotated as a J-domain protein named AtJ8 in Arabidopsis and as a truncated clone of pea PsJ8b Chiu et al , 2010 ).…”
