The purpose of this study was to investigate how nicotine in the context of waterpipe tobacco smoking (WTS) affects depression and anxiety-like behaviors associated with chronic social defeat stress (CSDS) in a male C57BL/6J animal model. The mice were exposed to WTS or received intraperitoneal injections of nicotine for thirty days, before being subjected to CSDS for ten days. During CSDS, mice were exposed to WTS or received nicotine injections. The social interaction and open-field tests were used to classify animals as resilient or susceptible to stress and to evaluate their anxiety-like behavior and social interaction. Mice post behavioral testing, were further exposed to WTS/nicotine for ten days, and their behavior was reassessed. The involvement of brain-derived neurotrophic factor signaling in the nicotine-mediated effects was assessed with the tropomyosin receptor kinase B (TRKB) inhibitor, ANA-12. The reported results indicated that WTS promoted resilience to stress and rescued social avoidance in the used mice model. Even though initially, WTS decreased anxiety-like behaviors it was found that prolonged exposure after the completion of CSDS significantly induced anxiety-like behaviors. Finally, we showed that nicotine mediates the effects of WTS only on resilience to stress by increasing BDNF and TRKB levels and signaling. Our results suggested that the pathways mediating resilience to stress and anxiety are distinct and that nicotine mediates the effects of WTS on social behavior, but not anxiety, by activating BDNF signaling.

Resistance training prevents right ventricle hypertrophy in rats exposed to secondhand cigarette smoke

Exposure to secondhand cigarette smoke is associated with the development of diverse diseases. Resistance training has been considered one of the most useful tools for patients with pulmonary disease, improving their quality of life. This study aimed to evaluate the effect of resistance training (RT) on the prevention of thickening of the right ventricle wall of rats exposed to secondhand cigarette smoke. Thirty-two Wistar rats were divided into four groups: Control (C), Smoker (S), Exercised (E) and Exercised Smoker (ES). The smoker groups were exposed to the smoke of four cigarettes for 30 min, twice daily, five days a week, for 16 weeks. The exercised groups climbed on a vertical ladder with progressive load, once a day, five days a week, for 16 weeks. The heart, trachea, lung, liver and gastrocnemius muscle were removed for histopathological analysis. Pulmonary emphysema (S and ES vs C and E, P < 0.0001) and pulmonary artery thickness enlargement (S vs C and E, P = 0.003, ES vs C, P = 0.003) were detected in the smoking groups. There was an increase in the right ventricle thickness in the S group compared with all other groups (P < 0.0001). An increase in resident macrophages in the liver was detected in both smoking groups compared with the C group (P = 0.002). Additionally, a relevant reduction of the diameter of the muscle fibers was detected only in ES compared with the C, S and E groups (P = 0.0002), impairing, at least in part, the muscle mass in exercised smoking rats. Therefore, it was concluded that resistance training prevented the increase of thickness of the right ventricle in rats exposed to secondhand cigarette smoke, but it may be not so beneficial for the skeletal muscle of smoking rats.

This study evaluated the behavioural responses of male Wistar rats to normal air or cigarette smoke (CS) and compared the effects of curcumin and hesperidin on growth performance. In experiment 1, male rats were randomised into two groups (n = 10): control and CS-exposed groups. During exposure (four weeks), the dietary and behavioural patterns were monitored. In experiment 2, forty-eight rats were distributed across eight groups (n = 6): normal control, CS control, CS + curcumin (10 mg.kg−1), CS + curcumin (20 mg.kg−1), CS + hesperidin (10 mg.kg−1), CS + hesperidin (20 mg.kg−1), curcumin (20 mg.kg−1), and hesperidin (20 mg.kg−1) for 6 weeks. Growth performance (feed intake, weight gain, and feed conversion ratio FCR) were assessed. In the first experiment, there was no significant difference (P > 0.05) in the body weight of the CS-exposed group compared to the normal control, whereas feed intake was significantly (P > 0.05) lower in the CS-group. The time to access feed and water was higher in the CS-group, while other behavioural responses (locomotion, stand upright, climbing, stand and stare, sniffing, sitting, and digging) were significantly reduced (P < 0.05) compared with normal control, especially after two weeks. In the second experiment, weight gain, feed in-take, and FCR were significantly lower in the CS-exposed group compared to the control group, whereas treatment with curcumin and hesperidin, especially at the higher dose (20 mg.kg−1 b. wt.), significantly improved the growth performance of the CS-exposed groups. This study submits that CS exposure negatively impacts on the growth performance and behavioural patterns and demonstrates the potentials of curcumin and hesperidin in addressing these CS-provoked changes.
