TNP1 Functional SNPs in bta-miR-532 and bta-miR-204 Target Sites Are Associated with Semen Quality Traits in Chinese Holstein Bulls1

Abstract: Transition nuclear proteins (TNPs), major proteins found in the chromatin of condensing spermatids, have been implicated in spermatogenesis and male fertility. In this study, DNA samples were collected from 404 Chinese Holstein bulls and sequenced to identify genetic variants in the 3'-untranslated region (UTR) of TNP1 and to investigate genetic variations in the TNP1 gene and their common haplotypes. This study was also conducted to determine whether these variations affect bovine semen quality traits and exp… Show more

“… 25 found that SIRT1 is the direct target gene of miR-204-5p 38 , and SIRT1 has been demonstrated to promote the expression of adipose triglyceride lipase gene and glycerol release 39 . Moreover, bta-miR-204 was determined to be present in the reproductive organs and was found to be significantly associated with semen quality traits in bovines 40 . These results indicated that bta-miR-204 may be involved in lipid metabolism in cattle and may be regulated by testosterone.…”

“…The relationship between the genotype or haplotype combinations of the INCENP gene and semen quality traits was analyzed using the least squares method as applied in the general linear models (GLM) procedure in SAS 9.0 (SAS Institute Inc., Cary, NC, USA). The GLM is given as follows [ 15 ]: where Y ijkl is the observed value of each semen quality trait, μ is the overall mean, G i is the fixed effect of genotype or haplotype combinations, P j is the fixed effect of age [j = 2–10; classified as (1) 2 years to 3 years; (2) 4 years to 5 years; (3)≥ 6 years], S k is the fixed effect of the origin of the bull, M l is the effect of the farm, and e ijkl is the random residual error. Values at P < 0 .…”

“…MicroRNAs are involved in multiple biological phenomena, such as spermatogenesis and early embryogenesis and tumorigenesis [ 12 , 13 , 14 ]. The SNPs in the binding site of miRNA, namely 3'UTR of the targeted mRNA, have been considered the principal element of microRNA adhesion and are associated with a phenotype, such as semen quality trait [ 15 ].…”

“…Zhang et al . 19 provided the first indication of g.442A > G-mediated and g.528G > A-mediated translational suppression in which SNPs alter the binding of bta-miR-204 and bta-miR-532 to the 3′-UTR of TNP1; the mediated translational suppression could be involved in the regulation of TNP1 expression and may influence the morphological characteristics of Chinese Holstein bull sperm. Liu et al .…”
