Background and aimsSeveral studies were conducted to explore the prognostic value of platelet-to-lymphocyte ratio (PLR) in pancreatic cancer and have reported contradictory results. This study aims to summarize the prognostic role of PLR in pancreatic cancer.Materials and methodsEmbase, PubMed and Cochrane Library were completely searched. The cohort studies focusing on the prognostic role of PLR in pancreatic cancer were eligible. The overall survival (OS) and progression-free survival (PFS) were analyzed.ResultsFifteen papers containing 17 cohort studies with pancreatic cancer were identified. The results showed patients that with low PLR might have longer OS when compared to the patients with high PLR (hazard ratio=1.28, 95% CI=1.17–1.40, P<0.00001; I2=42%). Similar results were observed in the subgroup analyses of OS, which was based on the analysis model, ethnicity, sample size and cut-off value. Further analyses based on the adjusted potential confounders were conducted, including CA199, neutrophil-to-lymphocyte ratio, modified Glasgow Prognostic Score, albumin, C-reactive protein, Eastern Cooperative Oncology Group, stage, tumor size, nodal involvement, tumor differentiation, margin status, age and gender, which confirmed that low PLR was a protective factor in pancreatic cancer. In addition, low PLR was significantly associated with longer PFS when compared to high PLR in pancreatic cancer (hazard ratio=1.27, 95% CI=1.03–1.57, P=0.03; I2=33%).ConclusionIn conclusion, it was found that high PLR is an unfavorable predictor of OS and PFS in patients with pancreatic cancer, and PLR is a promising prognostic biomarker for pancreatic cancer.

Background: Pancreatic carcinoma (PC) is one of the most aggressive cancers affecting human health. Identifying candidate biomarkers is essential for the early diagnosis and good prognosis of PC. Fibronectin type III domain containing 3B (FNDC3B) promotes many types of cancer, but its role in pancreatic cancer is not clear. The purpose of this study was to investigate the relationship between the expression of FNDC3B and incidence of PC. Methods: We downloaded data related to the levels of FNDC3B mRNA in patients of PC from the Cancer Genome Atlas (TCGA) database, and conducted in-depth research on this data and its clinical significance through the R tools. We used GO and KEGG enrichment analyses to study the genes and signaling pathways that may be regulated by FNDC3B. Results: The results showed that the level of FNDC3B mRNA in PC tissues was higher than that in the normal tissues, and this was associated with proliferation and lymph node metastases of PC. Increased levels of FNDC3B mRNA also predict poor prognosis for the patients with PC. In addition, enhanced levels of FNDC3B mRNA were involved in the regulation of cell junction, tissue and epithelial cell migration, and several signal transduction pathways, including notch, TGF-β, VEGF, and Wnt. Conclusions: In short, high levels of FNDC3B mRNA may be associated with progression of PC and indicate a poor prognosis in patients with PC.

Sex determining region Y-box 7 (SOX7) is known to function as a tumor suppressor in a number of types of cancer; however, its role in liver and pancreatic carcinoma remains unclear. The present study investigated the association between SOX7 expression and the clinical pathology of these carcinomas, in particular if SOX7 expression may be used to predict recurrence and patient prognosis following radical resection of liver and pancreatic carcinoma. SOX7 expression in human liver and pancreatic carcinoma was detected by immunohistochemical analyses and validated using mRNA data from a high-throughput sequencing dataset from The Cancer Genome Atlas (TCGA). SOX7 expression was significantly downregulated in liver and pancreatic carcinoma relative to the adjacent benign tissues [immunoreactivity scores: Liver carcinoma (3.53±1.57) vs. benign (7.00±0.00), P<0.001; and pancreatic carcinoma (2.39±1.88) vs. benign (4.80±0.45), P=0.005]. In addition, downregulation of SOX7 was significantly associated with advanced stage liver carcinoma, and the primary pathological tumor stage and regional lymph node stages. These findings were further validated in the TCGA dataset. However, SOX7 down regulation was closely associated with the only pathological grade in pancreatic patients. Kaplan-Meier analyses revealed significant differences in overall and disease-free survival between patients with high and low levels of SOX7 expression. In addition, a multivariate analysis with Cox regression indicated that SOX7 may be an independent predictor of disease-free survival. The results indicate that SOX7 may inhibit the progression of liver carcinoma and that SOX7 downregulation may accurately predict poor prognosis in liver carcinoma patients.
