TNF-αG-308A polymorphism is associated with insulin resistance: a meta-analysis

Abstract: ABSTRACT. Tumor necrosis factor-α (TNF-α) promoter polymorphisms has been reported to be associated with obesity and insulin resistance and gained widespread attention. However, results obtained so far are quite conflicting. We therefore performed a metaanalysis to address this issue, basing on 17 studies from electronic databases (MEDLINE and EMBASE). No evidence of significant effect of TNF-α G-308A polymorphism on body mass index (BMI) or obesity risk was detected (BMI: WMD RE = 0.05, 95%CI: -0.62 to 0.73; … Show more

“…Based on these data, the present meta-analysis showed no significant correlation between the TNF-α 308G/A polymorphism and PCOS. However, previous meta-analyses have demonstrated this variation to be associated with increased susceptibility to hepatocellular carcinoma, gastric cancer, and type 2 diabetes mellitus (Hu et al, 2014;Rokkas et al, 2014;Zhao et al, 2014;Wang, et al, 2015). Such inconsistent results may have been caused by the TNF-α 308G/A polymorphism performing divergent roles in different cell types or tissues.…”

“…Tumor Necrosis Factor-α (TNFα) is another key proinflammatory cytokine in the early stages of the inflammatory response ( 26 ). Similar to IL-1, elevated TNFα levels have been observed in crevicular fluid and saliva of patients with active peri-implantitis lesions and shown to directly correlate with the degree of clinical severity of peri-implant disease ( 27 ).…”

TNFα, among other direct and indirect effects, has a strong capacity to stimulate bone resorption by promoting cell differentiation from the monocyte-macrophage line to osteoclasts, inducing the maturation of osteoclasts and enhancing their resorptive activity ( 26 , 27 ).…”

“…Several polymorphisms have been identified in the promoter region of the TNFA gene [ 23 , 24 ]; however, to date, −308G → A polymorphism (rs1800629) has been the best characterized genetic variant in relation to metabolic and cardiovascular disease outcomes [ 24 26 ]. The TNFA -308G → A polymorphism has been linked with BMI and obesity [ 13 , 27 ], where A allele carriers have been shown to have higher BMI [ 27 ].…”
