“…Previous studies revealed that both the ACLfs and SCs are very susceptible to TNF-α in regulating the expressions of MMPs and LOXs. 8,14 TNF-α could destroy the balance of ECM through various ways, including inhibition of LOXs expression in the ACLf, promotion of MMPs expression in the ACLfs, and perisurrounded SCs. Previous data have proved that both ACLfs and SCs in the presence of TNF-α has high MMP activities, which have been hypothesized to primarily be responsible for the poor self-healing of cruciate ligaments in monoculture condition.…”

“…13 Previous studies have demonstrated that the ACLfs and synovial fibroblasts are very sensitive to TNF-α in regulating repair behavior of injured ACL. 14,15 However, very few effects are known to the TNF-α responsiveness of ACLfs in SCs' coculture condition.…”

“…In fact, there are some recent studies that points-out to the idea of the self-healing of the ACL breakage, although this capacity is probably limited (Fujimoto, Sumen, Ochi, & Ikuta, 2002;Kohl et al, 2013) and may be influenced by acute phase inflammatory mediators such as the tumor necrosis factor alpha (TNF-α) (Xie et al, 2014). The healing efficiency is mediated by the fibroblastic lysyl oxidase enzyme, which function is to catalyze the cross-linking between the collagen and the elastin and whose activity increases with the levels of TNF-α (Xie et al, 2014). As in our experiments, the dissection of the ACL and the repair was done in the same surgical procedure; this acute-phase healing mechanism above described could explain that both neo-formed ACL had the same macroscopic aspect.…”

“…Our previous studies elucidated that the IL-1 could increase LOX expressions in the ligament of human knee, 33 while the TNF-could decrease the LOX expressions. 34 The TGF-1 is an important growth factor in tissue remodeling that can enhance LOX expressions to promote the wound healing in ligaments. 35 In the present study we elucidated the expressions of LOXs induced by different dosages of the TNF-.…”
