TNF Receptor Family Member BCMA (B Cell Maturation) Associates with TNF Receptor-Associated Factor (TRAF) 1, TRAF2, and TRAF3 and Activates NF-κB, Elk-1, c-Jun N-Terminal Kinase, and p38 Mitogen-Activated Protein Kinase

Abstract: BCMA (B cell maturation) is a nonglycosylated integral membrane type I protein that is preferentially expressed in mature B lymphocytes. Previously, we reported in a human malignant myeloma cell line that BCMA is not primarily present on the cell surface but lies in a perinuclear structure that partially overlaps the Golgi apparatus. We now show that in transiently or stably transfected cells, BCMA is located on the cell surface, as well as in a perinulear Golgi-like structure. We also show that overexpression… Show more

“…Some members are established protein markers for this cell type and all are known to play a Immune response in silico AR Abbas et al a member of the TNF receptor family that is hypothesized to transduce signals for B-cell survival and proliferation. 43 Monocytes and macrophages Several distinct patterns of specific monocyte and macrophage expression are seen in Figure 3d and Supplementary Figure 3c. Expressed only in monocytes is PRAM1, an adaptor protein that appears to be involved in the differentation of monocytes.…”

“…BCMA -/-mice are born with no major immune defect apart from impaired survival of some plasma cells in the bone marrow [56]. It has been shown that BCMA binds to TRAF1, 2, and 3 in COS7 fibroblasts [64]. Experiments using TRAF1 -/-, TRAF2 -/-, and TRAF3 -/-plasma cells will be helpful in elucidating the functions of TRAFs in BAFF/BCMA-mediated plasma cell survival.…”

“…[14][15][16][17] The intracellular domains of BCMA and TACI associate with TNFreceptor-associated factors (TRAFs), leading to activation of NF-kB. 18,19 Recently, another receptor for BLyS, BAFF-R (also called BR3) was identified, which does not bind to APRIL. 20,21 Several lines of evidence indicate that these receptorligand systems crucially contribute to the pathogenesis of the autoimmune diseases.…”
