Tissue-Specific Distribution of iNKT Cells Impacts Their Cytokine Response

Abstract: Summary Three subsets of invariant natural killer T (iNKT) cells have been identified, NKT1, NKT2 and NKT17, which produce distinct cytokines when stimulated, but little is known about their localization. Here, we have defined the anatomic localization and systemic distribution of these subsets and measured their cytokine production. Thymic NKT2 cells that produced interleukin-4 (IL-4) at steady state were located in the medulla and conditioned medullary thymocytes. NKT2 cells were abundant in the mesenteric l… Show more

“…Studies in germ-free mice suggest that the iNKT cell compartment can be shaped by CD1d-lipid signals in the enteral tract (11,25), and thus early-life exposure to cow's milkbased infant formula might similarly shape human mucosal iNKT cells in early life. In adult mice, orally delivered antigen activates iNKT cells in the gut (26), suggesting that exposure to dietary lipid antigens in adult life may also have consequences. Interestingly, it is unclear whether cows have iNKT cells (27), as their CD1d does not present the long-chain α-galactosylceramide commonly used as a tool in mice and humans (28).…”

“…Although iNKT cell subsets preferentially secrete Th1, Th2, or Th17 cytokines, this response is not absolute given that many iNKT cells produce both IL-4 and IFN-g, and the extent of their activation is influenced by their location within lymphoid organs (53). Given that our set of TCRs induced different frequency of iNKT cell subsets, we predicted that they would also polarize the iNKT cytokine response upon Ag stimulation.…”

“…This imaging/analysis pipeline allows multiplex phenotypic identification and quantification of cells directly in tissues, akin to in situ flow cytometry. This technique has been used to study the composition, distribution, and function of densely packed cells with complex phenotypes and morphology in tissue sections (13)(14)(15)(16)(17)(18)(19)(20)(21). Even for cell types with relatively simple morphology, such as T and B lymphocytes, histo-cytometry requires a high degree of spatial signal resolution, achieved with high numerical aperture (N.A.)…”
