Tissue-specific analysis of lipid species in Drosophila during overnutrition by UHPLC-MS/MS and MALDI-MSI

Abstract: Diets high in calories can be used to model metabolic diseases, including obesity and its associated comorbidities, in animals. Drosophila melanogaster fed high-sugar diets (HSDs) exhibit complications of human obesity including hyperglycemia, hyperlipidemia, insulin resistance, cardiomyopathy, increased susceptibility to infection, and reduced longevity. We hypothesize that lipid storage in the high-sugar-fed fly’s fat body (FB) reaches a maximum capacity, resulting in the accumulation of toxic lipids in othe… Show more

“…Besides CO 2 , FlyNap, with an active ingredient of triethylamine, is another anesthetic commonly used to anesthetize Drosophila 51 . Unlike CO 2 or cold anesthesia, administration of triethylamine seizes all major motor activity in flies, but maintains normal respiratory function 52 . Respirometric measurements of flies anaesthetized using triethylamine provide a baseline rate of CO 2 production and metabolism of a fly without any major movement.…”

“…Mass spectrometry (MS) methods benefit from high sensitivity and requirement for low sample volumes and can be used in conjunction with isotope labeled nutrients and hormones. Combined HPLC and MS techniques were used to quantify tissue specific lipid accumulation ( Tuthill et al, 2020 ). Liquid chromatography tandem MS (LC-MS/MS) experiments can detect and quantify 13 C-labeled glucose in hemolymph drawn from flies reared on the labeled medium ( Cocuron et al, 2020 ).…”
