Tissue Recombination Models for the Study of Epithelial Cancer

Abstract: Animal models of cancer provide fundamental insight into the cellular and molecular mechanisms of human cancer development. As an alternative to genetically engineered mouse models, increasing evidence shows that tissue recombination and transplantation models represent an efficient approach to faithfully recapitulate solid epithelial cancer in mice. Cancer can be rapidly initiated through lentiviral delivery of defined genetic alterations into target cells that are grown in a physiological milieu with an appr… Show more

“…Much current understanding is drawn from experiments using mutant mice or immortalized cell lines or from investigation of established cancers in all of which models tumors must achieve a certain mass to be detected and hence to be sampled. In no model can one glimpse the moment a normal cell becomes a transformed cell or sample cells at the inception of cancer [ 67 , 68 , 69 , 70 ]. For this reason, there has been great enthusiasm (although much controversy as well) for validating the fundamental concepts of oncogenesis derived from experimental models and from analysis of tumor samples with cancer epidemiology [ 71 , 72 , 73 , 74 , 75 , 76 ].…”

“…While organoid culturing has the advantage of being able to manipulate single (ie, stromal) factors to assess their effect on tumor growth, this ex vivo approach has limitations. In vivo tissue recombination approaches could provide a more physiological context that better mimics the prostate microenvironment . Human prostatic epithelial cells can be recombined with human prostatic stromal cells and grafted under the renal capsules of mice.…”
